摘    要:以本研究室建立的恶性胶质瘤细胞系SHG-44为抗原,通过杂交瘤技术建立了两株恒定地分泌抗胶质瘤单克隆抗体的杂交瘤细胞株SZ-38、SZ-39。以该两株单克隆抗体为生物探针,在分子生物学水平研究了两种新的胶质瘤抗原,它们分别为胶质瘤细胞表面分子量47 000及180 000的糖蛋白。通过SZ-39对脑瘤内的放射免疫定位研究,确认了该单克隆抗体应用于临床肿瘤定位诊断和导向治疗的可能性。

Abstract:Hybridoma cells were derived by a fusion between NS-1 myeloma cells and spleen cells from a mouse immunized with human glioma cell line SHG- 44. After selective culture, screening and subcloning, two hybridoma lines (SZ-38, SZ-39) secreting McAb against the human glioma cells have been established. The specificity of these two McAb were then investigated on a panel of 10 cell lines derived from human glioma, 10 cell lines from other neoplasma cell lines, 7 different normal cells and 12 different human tissues and 50 brain tumors by ELISA, IIF and ABC method. The results indicated the antigens recognized by McAb SZ-38, SZ-39 may be gliomaassociated antigens. The SZ-38, SZ-39 were detected by Western blotting and immu-noprecipitation in molecular level.The SZ-38 was a Mw 47,000 and SZ-39 was a Mw 180,000 glycoprotein. Radio-immnnolocalization of ~(131)I-McAbSZ-39 was investigated in nude mice bearing human brain tumors by means of r-imaging, auto-radiography and radio-activity distribution.The results demonstrated the specificity of McAbSZ-39 in vivo and indi-cated that McAbSZ-39 may be used in clinical radio-immunolocalization and immunotreatment.
