摘    要:目的:考察保肝颗粒对四氯化碳(CCl4)致小鼠急性化学性肝损伤的保护作用。方法:90只昆明小鼠按体重随机分为正常组、模型组、护肝片组(0.7 g·kg-1)、保肝颗粒高、中、低剂量组(12.9,6.45,3.225 g·kg-1),每组15只。每日ig给药1次,连续30 d。末次给药1 h后,除正常组外,其余各组分别ip 0.1%CCl410 m L·kg-11次,建立小鼠急性化学性肝损伤模型,24 h后处死小鼠,收集肝组织及血清标本,检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和乳酸脱氢酶(LDH)活性;测定肝组织总超氧化物歧化酶(T-SOD)活性、还原型谷胱甘肽(GSH)和丙二醛(MDA)含量及进行肝组织病理学检查。结果:与正常组比较,模型组小鼠血清ALT,AST,LDH水平明显升高,肝组织T-SOD活性及GSH含量明显降低,MDA含量明显升高,均具有明显的统计学差异(P0.05),模型组肝组织病理学发现中央静脉和肝细胞结构变形明显,肝细胞排列紊乱,肝细胞肿胀、界限不清,细胞核变大,有的部位核仁消失,肝血窦变小、有充血,并可见大量成团的炎性细胞;与模型组比较,保肝颗粒高、中、低剂量组均能明显降低肝损伤小鼠血清ALT,AST,LDH水平(P0.05),提高肝组织T-SOD活性及GSH含量(P0.05),降低MDA含量(P0.05),并呈现量效关系;明显改善肝组织的病理学损伤。结论:保肝颗粒对CCl4致小鼠急性化学性肝损伤具有抗氧化及保护肝功能作用。

Abstract:Objective: To observe the protect effects of Baogan granules on acute chemical liver injury of mice induced by carbon tetrachloride (CCl4). Method: Ninety Kunming mice were randomly divided into normal group, model group, hugan tablet group(0.7 g·kg-1), baogan granules high, middle, low dose group(12.9, 6.45, 3.225 g·kg-1). Mices were given medicine once a day continuing 30 days. 1h after the last administration, in addition to the normal l group, each group respectively ip 0.1%CCl4 10 mL·kg-1, acute chemical liver injury in mice model was established. 24 h after death mice, collect liver tissue and serum specimens, detection of serum glutamic acid transaminase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) activity;determination of liver tissue total superoxide dismutase (T-SOD) activity, reduced glutathione (GSH) and malondialdehyde (MDA) content and hepatic histopathological examination. Result: Baogan granules can significantly reduce liver damage ALT, AST and LDH level in mice (P<0.05), Increase the T-SOD activity and GSH content (P<0.05), Reduce the MDA content in liver homogenate (P<0.05), and show concentration-response relationship;Greatly improve the pathology of liver tissue injury. Conclusion: Baogan granules on CCl4 induced acute chemical liver injury in mice has antioxidant and protect liver function function.
