摘    要:目的:优选白花蛇舌草中齐墩果酸和熊果酸的提取工艺,为该药材的资源开发提供参考。方法:在单因素试验基础上,以齐墩果酸和熊果酸提取量的综合评分为指标,根据Box-Benhnken中心组合设计原理,利用响应面分析法考察料液比、提取次数、提取时间对白花蛇舌草中齐墩果酸和熊果酸提取工艺的影响。结果:最佳提取工艺为加10.5倍量85%乙醇加热回流提取2次,每次92 min。齐墩果酸、熊果酸提取量分别为0.497 6,2.036 4 mg·g-1,综合评分97.23分,与预测值(99.05分)的相对误差1.8%。结论:通过响应面法优选的白花蛇舌草提取工艺合理可行,提取率高,易于工业化大生产,为该药材的开发应用提供实验依据。

Abstract:Objective: To optimize extraction process of oleanolic acid and ursolic acid from Hedyotis diffusa. Method: On the basis of single factor tests, according to Box-Behnken design principles, response surface analysis was adopted to investigate effects of solid-to-liquid ratio, extraction times and time on comprehensive score of extracting amounts of oleanolic acid and ursolic acid from H. diffusa. Result: The best extraction process of oleanolic acid and ursolic acid was adding 10.5 times the amount of 85% ethanol, reflux extracting for twice, 92 minutes for each time.Yield of oleanolic acid and ursolic acid were 0.497 6 mg·g-1 and 2.036 4 mg·g-1, respectively;comprehensive score was 97.23 points, its relative error was 1.8% by comparing with the predicted value. Conclusion: This process is reasonable and feasible with high extraction rate, and it is suitable for industrial production.This study can provide experimental basis for further development and application of H. diffusa.
