补肾法对去势兔动脉粥样硬化原代培养平滑肌细胞凋亡及血小板源生长因子A mRNA等表达的影响

摘    要:目的:观察补肾煎对去势兔动脉粥样硬化原代培养平滑肌细胞凋亡及血小板源生长因子A(PDGF-A) mRNA与单核细胞趋化蛋白1(MCP-1) mRNA表达的影响.方法:新西兰雌兔随机分为A组(卵巢切除)、B组(未切除卵巢)、C组(卵巢切除 补肾煎),均予高脂饮食并注射胎牛血清蛋白,造成动脉粥样硬化模型.同时C组予补肾煎,12周后检测相关指标.结果:C组培养平滑肌细胞凋亡率高于A组、B组, PDGF-A mRNA 及MCP-1 mRNA在A组、C组的表达明显高于B组(P<0.01),而在C组的表达又显著低于A组(P<0.01).结论:补肾煎可能通过增加血管平滑肌细胞的凋亡、降低PDGF-A mRNA及MCP-1 mRNA的表达而减轻去势兔动脉粥样硬化的形成.

Abstract:Objective: To observe effects of Bushen Decoction on apoptosis of primary cultured smooth muscular cells and expressions of platelet derived growth factor-A (PDGF-A)mRNA and monocyte chemotactic protein-1 (MCP-1)mRNA. Methods:New Zealand female rabbits were randomly divided into group A(ovariectomy), group B(ovary not removed) and group C(ovariectomy plus Bushen Decoction). They were all given high fat forage and injection of bovine serum protein to develop model of atherosclerosis. At the same time, Bushen Decoction was given to group C. The relative indexes were detected 12 weeks later. Results: The apoptosis rate of smooth muscular cells in group C was higher than those in group A and B; The expressions of PDGF-A mRNA and MCP-1 mRNA in group A and C were higher than those in group B(P<0. 01), and in group C was significantly lower than that in group A(P<0. 01). Conclusion: Bushen Decoction can reduce formation of atherosclerosis in ovariectomized rabbit by increasing apoptosis of vascular smooth muscular cells and down-regulating expressions of PDGF-A mRNA and MCP-1 mRNA.
