摘    要:目的 通过生物信息学方法筛选慢性肾脏病(chronickidneydisease,CKD)可能的自噬关键风险基因,同时预测治疗CKD的潜在中药药物,探讨致病的潜在靶点及治疗药物。方法 从基因表达总表(Gene Expression Omnibus,GEO)数据库获取CKD患者肾组织测序数据。采用生物信息学方法提取正常对照组与CKD患者组的自噬相关差异表达基因(differentially expressed autophagy-related genes,DEARGs),并进行基因本体(gene ontology,GO)功能分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析。通过支持向量机模型(support vector machine,SVM)确定关键基因。最后,通过Connectivity Map平台对关键基因筛选可能的治疗CKD的中药。结果 共获得67个自噬相关差异基因,GO、KEGG富集分析表明DEARGs主要与巨自噬、自噬体、泛素样蛋白连接酶结合等相关。并从2个(ATF6、GNAI3)关键基...

Bioinformatics-based screening of key genes for autophagy in the progression of chronic kidney disease and prediction study of potential Chinese medicine
Abstract:Objective Screening potential autophagy key risk genes for chronic kidney disease (CKD) through bioinformatics methods, predicting potential traditional Chinese medicine drugs for treating CKD, and exploring potential pathogenic targets and therapeutic drugs. Methods In this study, renal tissue sequencing data of CKD patients were obtained from the Gene Expression Omnibus (GEO) database. Bioinformatics methods were used to extract autophagy-related differentially expressed genes (DEARGs) from normal control and CKD patient groups, and gene ontology (GO) functional analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed. Key genes were identified by support vector machine model (SVM). Finally, the key genes were screened for possible therapeutic Chinese medicines by the Connectivity Map platform. Results A total of 67 autophagy-related differential genes were obtained, and GO, KEGG enrichment analysis showed that DEARGs were mainly associated with macroautophagy, autophagosome, and ubiquitin-like protein ligase binding. And from 2 (ATF6, GNAI3) key genes screening, we obtained several herbal medicines with potential therapeutic effects, such as turmeric, zedoary, salvia, dioscorea and schisandra. Conclusion Through bioinformatics analysis, key autophagy genes and potential therapeutic traditional Chinese medicine have been identified in the progression of CKD, providing new ideas for the clinical diagnosis and treatment of CKD.
