| 摘 要： | 目的 采用生物信息学和分子生物学技术相结合,寻找在系统性红斑狼疮(SLE)中特异性表达的微小RNA,为进一步研究微小RNA在SLE发病机制中的作用打下基础.方法 查找与SLE发病相关的基因,采用微小RNA靶基因预测数据库预测SLE相关基因上的可能微小RNA靶位点,选择其相对应的微小RNA-146a,即hsa-miR-146a,采用实时荧光定量PER方法检测SLE患者与正常人外周血单个核细胞(PBMCs)中微小RNA的表达差异.结果 SLE患者外周血PBMCs中hsa-miR-146a的循环阈值的差值高于正常对照组(4.52±1.18比2.76±1.38,P=0.02),hsa-miR-146a的表达量显著低于正常对照组.结论 SLE患者PBMCs中hsa-miR-146a的表达水平降低,表明hsa-miR-146a在SLE发病机制中可能发挥一定的作用.
