脓毒症大鼠肾脏AQPs动态变化与水代谢的关系

摘    要:目的:探讨肾脏AQPs的表达与肾损伤水代谢紊乱的关系。方法:Wistar大鼠采用盲肠结扎穿孔法(CLP)建立脓毒症大鼠模型。观测尿量、尿渗透压和肾功能。免疫组化方法检测AQP1、AQP2蛋白水平,实时荧光定量PCR方法检测AQP1mRNA、AQP2mRNA水平。结果:手术组大鼠肾脏AQP1蛋白及AQP1mRNA表达随着CPL术后时间的延长呈下降趋势,6h开始下降,24h达最低;AQP2蛋白及AQP2-mRNA表达均在术后3h增高、12h、24h明显降低,较假手术组差异有统计学意义(P〈0.05)。大鼠肾脏AQP1、AQP2分别与尿量呈负相关(P〈0.05),与尿渗透压呈正相关(P〈0.05)。结论:AQP1和AQP2基因通过调节AQP1、AQP2蛋白的表达,参与肾脏水代谢的调节;AQP1、AQP2蛋白及基因的表达异常是导致脓毒症肾损伤水代谢紊乱的重要原因。

Abstract:Objective:To investigate the relation between the expression of renal AQPs and water metabolism disorder in AKI.Methods:Septic models of Wistar rats were established by cecum ligation perforation(CLP).The urine volume,urine osmotic pressure and renal function were observed.The expression of AQP1and AQP2 protein in rats' kidneys were detected by immunohistochemistry method,the expressions of AQP1 mRNA,AQP2 mRNA by real-time PCR.Results:In septic models,AQP1 protein and AQP1-mRNA expression decreased gradually after CLP.Began to decrease at 6 h,reaching the lowest at 24h (P0.05).The expression of AQP2 protein and AQP2-mRNA increased significantly at 3 h then decreased significant at 12 h and 24 h(P0.05).Renal AQP1,AQP2 were negatively correlated with urine volume in septic models group,respectively(P0.05),and were positively correlated with urine osmotic pressure respectively(P0.05).Conclusion:AQP1 gene and AQP2 gene were involved in the water metabolism mechanism of kidney by regulating the expression of AQP1 and AQP2 protein.The abnormal expression of protein and gene of AQP1 and AQP2 is an important cause of water metabolism disorders in septic kidney.
