免疫抑制剂预处理对同种异体气管移植犬免疫耐受的影响

摘    要:目的:诱导受体犬对经免疫抑制剂预处理的供体犬移植气管形成免疫耐受。 方法:实验用犬20只,随机选择4只做供体,其中2只经预处理犬(实验组供体)(术前6 h给予环磷酰胺750 mg、甲基强的松龙2 g静脉注射)的气管切成8段,每段长3 cm,移植入实验组受体内不用免疫抑制治疗;另外2只未经预处理犬(对照组供体)8段气管移植入对照组受体内。其余16只犬随机均分至2个受体组,分别将未经和经免疫抑制剂预处理的供体犬气管段移植到对照组和实验组受体犬腹腔大网膜包埋3周后取出,行病理学检查两组气管移植术后免疫排斥反应。 结果:实验组8段包埋气管中,除1段出现重度排斥反应外均为轻度排斥反应;对照组8段包埋气管中,3段出现轻度排斥反应,3段中度排斥反应,2段重度排斥反应;两组间气管移植术后免疫排斥反应比较差异无显著性(P>0.05)。 结论:经免疫抑制剂预处理的供体,能诱导受体对同种异体移植的气管形成免疫耐受;供体预处理与未经预处理而术后常规应用免疫抑制剂治疗有相同的效果。

Abstract:Objective To induce the immune tolerance of recipient to donor trachea pretreated with immunosuppressor. Methods Twenty adult dogs were used, 4 dogs were donors, the others were recipients. The donor dogs were killed and all tracheas were divided into 16 segments. The 2 donor dogs with 8 tracheal segments pretreated by immunosuppressor (CTX 750 mg and MPED 2 g) were used as experimental group, the other 2 donor dogs with 8 tracheal segments without immunosuppressor pretreatment as control group. After the tracheal segments were embedded with omentum in the recipient peritoneal cavity for 3 weeks , all tracheal segments were taken out to evaluate the scale of immune reaction pathologically. Results The normal histological construction was kept in 7 tracheal segments except 1 segment with severe immune rejection in the experimental group. 3 segments with mild rejection, other 3 segments with moderate rejection and another 2 tracheal segments with severe rejection were found in the control group. There was no significant difference between the two groups (P>0.05).Conclusion The pretreatment of donor with immunosuppressor can induce recipient immune tolerance for the canine tracheal allotransplantation.
