丙泊酚对大鼠冷缺血心肌的保护效应

摘    要:目的 观察丙泊酚不同处理方式对离体大鼠缺血再灌注心肌的保护效应,探索其最佳给药途径.方法 对大鼠离体心脏行改良Langendorff灌流,将32只SD大鼠随机分为4组,每组8只.A组持续正常K-H液灌注40min后全心停灌30min,再灌注60min;B组平衡20min后用含50/μmol/L丙泊酚K-H波灌注10min,再用K-H液冲洗10min,缺血与再灌注处理同A组;C组K-H液灌注40min,全心停灌30min后再灌注时先用含50μmol/L丙泊酚K-H液灌注10min,再用K-H液灌注50min;D组平衡20min后用含50μmol/L丙泊酚K-H液灌注20min,全心停灌30min,再用含50μmol/L丙泊酚K-H液灌注60min.观察缺血再灌注期间HR,LVDP、LVEDP、±dp/dtmax变化及灌注末心肌组织中丙二醛(MDA)含量、心肌超氧化物歧化酶(SOD)活性.结果 再灌注后,与A组相比,B、C、D组HR、LVDP、LVEDP、±dp/dtmax均有显著变化(P<0.01),D组各项血流动力学指标均优于B、C组(P<0.05).与A组相比,B、C、D组SOD活性均明显增高,MDA含量减少,差异有统计学意义(P<0.01),以D组更为显著.结论 丙泊酚对离体大鼠缺血再灌注心肌有保护效应,缺血前后持续以含50μmol/L丙泊酚K-H液灌注保护效应最为明显.
