诱导血红素氧合酶-1对小肠移植缺血再灌注损伤的影响

摘    要:目的:探讨诱导血红素氧合酶-1(HO-1)是否可减轻大鼠小肠移植缺血再灌注损伤及其机制。方法:SD大鼠分成3组:Ⅰ组:血晶素(Hemin)组(n=20);Ⅱ组:Hemin+卟啉锌(ZnPP)组(n=20);Ⅲ组:生理盐水组(n=20)。建立小肠移植缺血再灌注损伤模型。移植后6、12、24、48h分别从各组随机取5只大鼠,切取移植小肠进行HO-1活性测定.普通病理学检查及细胞凋亡检测。结果:术后各时段Ⅰ组HO-1活性均显著强于Ⅱ组及Ⅲ组(P〈0.05),而Ⅱ组和Ⅲ组比较无显著差异(P〉0.05)。各组移植肠缺血再灌注损伤以术后12h最为明显。Ⅰ组表现轻度,而Ⅱ组和Ⅲ组表现中度缺血再灌注损伤。术后各时段Ⅰ组细胞凋亡数少于Ⅱ组和Ⅲ组,差别有统计学意义(P〈0.05),而Ⅱ组和Ⅲ组比较无统计学意义(P〉0.05)。结论:诱导HO-1可通过抑制细胞凋亡而减轻移植肠缺血再灌注损伤。

Abstract:Objective:To investigate whether induced HO-1 could alleviate the ischemia-reperfusion injury in transplanted small bowel and to investigate the mechanism.Methods:SD rats were divided into three groups:group 1(hemin group,n = 20),group 2(hemin ZnPP group,n = 20)and group 3(saline group,n = 20).Ischemia-reperfulion injury model of small bowel transplantation was established.Five rats of each group were sacrificed 6,12,24 and 48 h after transplantation,then the activity of HO-1 in the tissue of transplanted intestine was detected,histological changes observed and apoptosis detected.Results:The post-operative activity of HO-1 in group 1 was significantly higher than that in group 2 and group 3(P < 0.05),but there was no significant difference in the activity of HO-1 between group 2 and group 3.All rats showed the most serious ischemia-reperfusion injury 12 hours after reperfusion,and the damage was milder in group 1 than that in the other two groups.At the same time,there were less apoptosis cells in group 1 than group 2 and group 3(P < 0.05).Conclusion:HO-1 can alleviate the ischemia-reperfusion injury of transplanted small bowel by reducing apoptosis.
