霍山石斛不同提取物对肾阴虚证、肾阳虚证小鼠抗疲劳作用及物质基础研究

摘    要:目的:研究霍山石斛水提物、醇提物对肾阴虚证、肾阳虚证小鼠抗疲劳作用及其物质基础。方法:180只健康昆明雄性小鼠适应性喂养7d后随机分为空白组、肾阴虚模型组、肾阳虚模型组、西洋参组、人参组、肾阴虚各给药组、肾阳虚各给药组,分别每日灌服甲状腺素溶液和氢化可的松溶液制备肾阴虚证、肾阳虚证动物模型,给药14d,检测各组小鼠的力竭游泳时间、肝糖原、肌糖原、血乳酸(BLA)、游离脂肪酸(FFA)、血尿素氮(BUN)、甘油三酯(TG)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、Na+-K+-ATP酶和Ca2+-ATP酶的变化。结果:与肾阴虚模型组比较,霍山石斛水提物各剂量组、醇提物高剂量组能升高模型小鼠的力竭游泳时间,升高肝糖原和肌糖原含量,降低血清FFA、BUN和TG,提高LDH值,降低肝脏CK、Na+-K+-ATP酶及Ca2+-ATP酶值(P<0.05,P<0.01)。与肾阳虚模型组比较,霍山石斛水提物高剂量组能升高模型小鼠的力竭游泳时间,升高肝糖原和肌糖原含量,降低BLA水平,升高血清FFA、BUN、TG、CK及肝脏LDH、Na+-K+-ATP酶值(P<0.05,P<0.01),霍山石斛醇提物高剂量能升高模型小鼠的力竭游泳时间和肌糖原含量(P<0.05)。结论:霍山石斛水提物能提高肾阴虚证、肾阳虚证小鼠的抗疲劳作用,具有双向调节作用,可能是霍山石斛补虚作用的主要物质基础,霍山石斛醇提物仅能提高肾阴虚证小鼠的抗疲劳作用,可能是霍山石斛滋阴清热功效的主要物质基础。

Abstract:Objective:To explore the anti-fatigue effect and material basis of different extracts of Dendrobium huoshanense on mice with kidney-yin deficiency and kidney-yang deficiency.Methods:A total of 180 healthy Kunming male mice were divided into control group,kidney-yin deficiency model group,kidney-yang deficiency model group,American ginseng group,ginseng group,administration groups of kidney-yin deficiency and kidney-yang deficiency after 7 days of adaptive feeding.The rats were fed with thyroxine solution and hydrocortisone solution daily to prepare animal models of kidney-yin deficiency and kidney-yang deficiency.After 14 days of administration,the changes of exhaustive swimming time,liver glycogen,muscle glycogen,BLA,FFA,BUN,TG,CK,LDH,Na+-K+-ATPase and Ca2+-ATPase were detected.Results:Compared with the kidneyyin deficiency model group,the low and high dose groups of Dendrobium huoshanense water extract and the high dose groups of Dendrobium huoshanense alcohol extract could increase the exhaustive swimming time,the level of liver glycogen and muscle glycogen,decrease the level of serum FFA,BUN and TG,enhance the activity of LDH in liver,decrease the activity of CK in serum,Na+-K+-ATPase,Ca2+-ATPase in liver(P<0.05,P<0.01).Compared with the kidney-yang deficiency model group,the high dose groups of Dendrobium huoshanense water extract could increase the exhaustive swimming time,the level of liver glycogen and muscle glycogen,decrease the level of serum BLA,enhance the levels of FFA,BUN,TG and CK in serum,the activity of LDH,Na+-K+-ATPase in liver(P<0.05,P<0.01).And the high dose groups of Dendrobium huoshanense alcohol extract could increase the exhaustive swimming time and the level of muscle glycogen(P<0.05).Conclusion:The water extract of Dendrobium huoshanense can improve the anti-fatigue effect of kidney-yin deficiency syndrome and kidney-yang deficiency syndrome in mice,and has two-way regulation effect.It may be the main material basis of the tonifying effect of Dendrobium huoshanense.The alcohol extract of Dendrobium huoshanense can only improve the anti-fatigue effect of kidney-yin deficiency syndrome in mice,which may be the main material basis of the nourishing yin and clearing heat effect of Dendrobium huoshanense.
