细辛脂素抗心脏移植急性排斥反应的作用及对黏附分子表达的影响

摘    要:目的:实验筛选细辛免疫抑制作用的有效成分细辛脂素,探讨其抗心脏移植急性排斥反应作用及对黏附分子的影响。方法:提取细辛中的细辛脂素,健康雄性Wistar大鼠(供体)和SD大鼠(受体)各64只,建立大鼠异位心脏移植模型,将接受心脏移植的SD大鼠分成4组:阳性对照组,不给任何处置;环孢霉素A(CsA)组,术前1 d开始灌服CsA;细辛脂素组,术前1 d开始灌服细辛脂素;混合药物组,术前1 d开始灌服半量CsA和半量细辛脂素。每组各8只,术后第7天处死,另外8只用于观察移植物生存天数。移植心常规石蜡包埋、切片,显微镜观察并进行组织病理分级;免疫组化染色法检测移植物局部细胞间黏附分子-1(ICAM-1)和血管内皮黏附分子-1(VCAM-1)含量。结果:移植心存活时间,CsA组、细辛脂素组和混合药物组均较阳性对照组显著延长(P<0.01)。与阳性对照组比较,细辛脂素可减轻心肌病理形态学的损伤。CsA组、细辛脂素组和混合药物组移植心局部ICAM-1和VCAM-1含量较阳性对照组明显减少。结论:细辛脂素可延长移植物存活时间并减轻病理形态的损伤,与CsA有相似的免疫抑制作用。ICAM-1和VCAM-1在发生排斥反应时表达增加,细辛脂素可抑制二者的表达。

Abstract:Objective: To test the effect of asarinin,the extract of Herba Asari,on the acute heart transplantation rejection and the expression of adhesion molecule.Method: Asarinin was extracted from herba asari.64 SD rats undergone heart transplantation were divided into four groups: group A(control group),group B (Cyclosporine A treated),group C(Asarinin treated),and group D(1/2 CsA and 1/2 Asarinin).Some rats were used to examine survival time(n=8) and the others were used to observe the pathological injury and the expression level of intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) by using immunohistochemistry.Result: Asarinin could prolong the survival time of allografts,which was similar to CsA group(P>0.05).Asarinin could relieve the damage of cardiomyocytes of the transplanted.Asarinin could also decrease the level of ICAM-1 and VCAM-1 in the allografts.Conclusion: Asarinin may play important roles in suppressing the immune rejection,prolong the allografts survival time and protect the donor organ,which was similar to CsA.The expression level of ICAM-1 and VCAM-1 is increased in suppressing the course of acute rejection and asarinin can inhibit their expression level.Asarinin can decrease the dosage of CsA.
