摘    要:目的:研究组织工程皮肤培养液中血管内皮细胞生长因子(VEGF)的分泌情况。方法:收集不同时相的单层成纤维细胞培养液、胶原凝胶真皮替代物(胶原凝胶DE)、胶原凝胶皮肤替代物(胶原凝胶SE)、复方壳多糖真皮替代物(复方壳多糖DE)、复方壳多糖皮肤替代物(复方壳多糖SE)培养液样本,双抗体夹心EUSA法测定其中VEGF分泌量。结果:成纤维细胞(FB)在三维培养时分泌的VEGF明显高于其单层培养时的分泌量。复方壳多糖组织工程皮肤VEGF的分泌量又高于胶原凝胶组织工程皮肤。大多数培养液中VEGF含量在48h内增长最快。结论:壳多糖组织工程真皮和皮肤能分泌较多的VEGF,有利于伤口的血管化,加速伤口愈合。

Abstract:Objective:To study the secretion of vascular endothelial growth factor(VEGF)in cultured engineering skin equivalent.Methods:The VEGF was detected by ELISA(antibody-VEGF-antibody)in the cultured medium which was collected at different time point from the cultures of monolayer fibroblast(FB),collagen gel dermal equivalent,collagen gel skin equivalent,composite chitosan dermal equivalent or composite chitosan skin equivalent.Results:The VEGF secreted by FB in three-dimensional culture was more than that in monolayer culture.Further more,the VEGF secreted by composite chitosan skin equivalent was more than that of collagen skin equivalent.VEGF has been shown to increase rapidly within 48 hours of culture.Conclusions:Composite chitosan skin equivalent,including composite chitosan dermal equivalent,has the ability of secreting more VEGF,which is contribute to the vascularization and healing of wounds.
