非酒精性脂肪性肝病大鼠肝组织内脂素的表达变化

目的观察非酒精性脂肪性肝病大鼠肝脏脂质沉积与内脂素因子的动态变化.方法随机将80只SD大鼠分成普通饲料和高脂饲料喂养组,两组按时间节点再分为4个亚组（各10只）.检测大鼠血清内脂素水平和肝组织内脂素蛋白及mRNA水平的变化.结果在高脂饮食喂养4周后,大鼠肝细胞出现轻度脂肪变,血清和肝组织内脂素mRNA水平分别为61.23±8.07μg/ml和5.36±1.91×10^3拷贝,与正常组59.57±4.57μg/ml和1.33±0.29×10^3拷贝比,无统计学差异;第9周时分别升高为67.53±11.46μg/ml和7.12±2.28×10^3拷贝（P〈0.05）,第13周时为76.35±14.05μg/ml和19.9±3.74×10^3拷贝（P〈0.01）,第18周实验结束时为81.89±13.62μg/ml和26.67±6.87×10^3拷贝（P〈0.01）.结论非酒精性脂肪性肝病大鼠血清及肝组织内脂素因子表达上调,提示其在非酒精性非脂肪性肝病发生发展过程中起一定的作用.

The change of serum and hepatic visfatin expression in rats with non-alcoholic fatty liver disease

Objective The aim of this study was to observe the dynamic change of serum and hepatic visfatin expression in rats with non-alcoholic fatty liver disease （NAFLD）. Methods 80 Sprague Dawley rats were divided into two groups with normal or high-fat diet feeding. Each group was further divided into four subgroup （n=10）. Serum visfatin was measured by ELISA,and visfatin levels in liver was detected with Western blotting and real time fluorescence quantitative PCR. Results After 4 weeks,the liver tissues in model rats showed steatosis while serum visfatin increased to 61.23±8.07μg/ml and high visfatin protein/mRNA expression could be observed; The serum visfain level reached to 67.53±11.46μg/ml and the visfatin mRNA were 7.12±2.28×10^3 copies in liver tissue （P〈0.05）;At thirteenth week,serum visfatin level was 76.35±14.05μg/ml,while its mRNA in liver tissues was 19.9±3.74×10^3 copies;At eighteenth week, serum visfatin was 81.89±13.62μg/ml and its mRNA was 26.67±6.87×10^3 copies （P〈0.01）. Conclusion The gradual up-regulation of visfatin expression in livers of rats with NAFLD indicates its certain role during the development of NAFLD.
