重组人p53腺病毒联合5-氟尿嘧啶对胃癌细胞的作用

目的 研究重组人p53腺病毒增加胃癌细胞对5-氟尿嘧啶（5-Fu）敏感性的作用。方法 重组人p53腺病毒感染胃癌细胞BGC-823，Western blot法检测p53蛋白在胃癌细胞中的表达；MTT法测定重组人p53腺病毒单独及联合5-Fu用药的不同浓度处理细胞的生长抑制率，流式细胞仪检测细胞周期分布和凋亡。结果 重组人p53腺病毒感染BGC-82348h，p53蛋白在BGC-823中高表达，并产生G2／M期阻滞和细胞凋亡。重组人p53腺病毒联合5-Fu用药增加5-Fu的敏感性，有剂量时间的依赖性。结论 腺病毒介导p53基因感染BGC-823细胞诱导凋亡并增加胃癌细胞对5-Fu的敏感性。本实验为p53基因治疗与胃癌化疗临床结合提供了可靠的实验依据。

Effect of adenovirus mediated p53 gene with fluorouacil to gastric cancer cells

Objective To evaluate the enhanced sensitivity of gastric cancer cells to fluorouacil by adenovirus mediated p53.Methods Recombinant adenovirus mediated p53 gene(rAd-p53)was transfected into human gastric cancer cells with unknown p53 genetic status.P53 protein expression was detected by Western blot assay.Cell survival and apoptosis of the gastric cancer cells was assessed by MTT assay.Cell cycle distribution and apoptotic rate were determined by flow cytometry.Results Inducing of G2/M arrest and apoptosis and inhibition of tumor cell proliferation were observed after the infection of adenovirus mediated p53,and efficiently p53 protein expression was demonstrated in the human gastric cancer cells in this study.Adenovirus mediated p53 could enhance the sensitivity of gastric cancer cells to Pacli5-FUel with the dependence of time and dose.Conclusion This study demonstrated that Adp53 transfer increased the cellular apoptosis and sensitivity of human gastric cancer cells in vitro,and supported that the combination of p53 gene therapy with chemotherapy may enhance the adenovirus mediated p53 on chemosensitivity of gastric cancer cells.
