摘    要:目的 研究P16基因改变与脑胶质瘤恶性程度分级及肿瘤细胞增殖活性之间的关系。方法 检测41例不同级别脑胶质瘤标本和7例正常脑组织P16基因改变和PCNA蛋白表达情况。结果 实验组P16基因缺失和突变22例(54%),mRNA和蛋白表达缺失分别为23例(56%)和27例(66%),三者改变非一对一关系。P16基因改变和PCNA指数随脑胶质瘤恶性程度级别增高呈上升趋势(P〈0.05)。P16基因改变与

Abstract:Objective To study the relationship between P16 gene modification and the malignant grades of brain glioma and the proliferative activity o f tumor cells.Methods The P16 gene modification and PCNA protein expression were detected in 41 brain glioma specimen with different grades and 7 normal brain tissue specimen. Results In the experimental group, the deletion or mutation of P16 gene were found in 54%(22/41) cases, the deletions of mRNA and protein expression were found in 56%(23/41) and 66%(27/41) cases respectively. The changes in DNA, RNA and protein levels were without one to one relation. The P16 gene modification and increase of PCNA index were related to the increase of malignant grade of brain glioma significantly (P<0 05). Also the P16 gene modification was related to the change of PCNA index dominantly (P<0 01). Conclusions The abnormity of cell cycle modulation and increase of cellular proliferation activity caused by deletion or mutation of P16 gene is an important factor to influence the biological characteristics of brain glioma.
