蛇纹石除氟砖茶水对大鼠DNA影响的实验研究

摘    要:目的观察蛇纹石除氟砖茶水对大鼠血、肝、肾单个核细胞DNA损伤的影响。方法用活化蛇纹石将含氟为5.0、50.0 mg/L的砖茶水分别除氟至1.5 mg//L,予以Wistar大鼠自由饮用,同时设立对照组和未除氟组,共分5组,计70只喂养3个月。用单细胞凝胶电泳技术观察血、肝、肾单个核细胞DNA百分含量、尾长和尾动量。结果除氟组血、肝、肾3种单个核细胞DNA损伤与对照组比较,差异均无统计学意义(P>0.05);含氟5.0、50.0 mg/L砖茶水组血单个核细胞DNA损伤与对照组、除氟组比较,差异均有统计学意义(P<0.05);含氟50.0 mg/L砖茶水组大鼠肝细胞DNA损伤与对照组、除氟组比较,差异均有统计学意义(P<0.05);砖茶水组肾单个核细胞DNA损伤与对照组、除氟组比较,差异无统计学意义(P>0.05)。结论未发现活化蛇纹石除氟砖茶水(含氟量1.5 mg/L)引起大鼠DNA损伤。

Abstract:Objective To observe the effect of serpentine-defluoridized brick-tea water on deoxyribonucleic acid (DNA) damage of single-nucleic cell from blood, liver, kidney of rats. Methods Fluorine ion contents in brick-tea water from 5.0,50.0 mg/L were decreased to 1.5 mg/L by activated serpentine,respectively. Wistar rats were divided into A, B, C, D and E groups according to fluorine ion concentrations (A. control group; B. 5.0 mg/L to 1.5 mg/L; C. 50.0 mg/L to 1.5 mg/L; D. 5.0 mg/L; E. 50.0 mg/L). All the rats were fed with different fluorine ion concentrations of brick-tea water for 3 months. Single-nucleic cells in blood, liver and kidneys underwent single cell gel electrophoresis technique. If DNA damage has occurred the electrophoresed DNA fragments appear as a diffuse tail behind the nucleus known as a "comet". The length and the motion of the tail and the amount of DNA damage was analyzed. Results Compared with the control group (group A), DNA indicators (tail DNA, length and ext-mom) in single nucleic cell in blood, liver and kidneys had no significant differences in group B, C(P > 0.05). There were significant differences of DNA indicators in single liver cell among group E and group A,B,C (P < 0.05). Tail DNA,length and ext-mom in group E was higher that those in group A,B,C (P < 0.05). In comparison with group A,B,C, DNA indicators in single nucleic cell in blood have significant differences(P< 0.05), but not in kidneys in group D,E (P > 0.05). Conclusions Defluoridized brick-tea water containing 1.5 mg/L fluorine ion concentrations by activated serpentine does not lead to DNA damage in rats.
