p63,p73蛋白在乳腺癌组织中的表达及其意义

摘    要:目的:探讨p63和p73蛋白在乳腺组织的表达情况及其与乳腺癌生物学行为的关系。方法:用免疫组化SP法检测5例正常乳腺组织(对照组),5例增生乳腺组织(增生组),15例原位乳腺癌组织(原位组),60例浸润性乳腺癌组织(浸润组)中p63蛋白、p73蛋白的表达。结果:乳腺良性病变的腺泡和导管周围可见连续的肌上皮围绕,p63阳性;原位组p63染色显示肌上皮呈不连续表达;乳腺癌早期浸润灶及浸润性癌,p63染色显示肌上皮大部分消失或无肌上皮。p73在对照组、增生组、原位组及浸润组中的阳性率分别为0%(0/5),20.0%(1/5),33.3%(5/15)和66.7%(40/60),差异有显著性(P〈0.05);在乳腺肿瘤组织中,随组织学病理分级的增加、临床分期的进展和腋窝淋巴结的转移,p73表达阳性率则逐渐上升,差异有显著性(P〈0.05);p63蛋白、p73蛋白表达无相关性。结论:p63、p73在乳腺癌的发生、发展过程中起着重要的作用;二者联合检测可为乳腺癌的早期诊断、及时治疗及预后判断提供必要的理论依据。

Abstract:Objective:To investigate the expression of p63 protein and p73 protein in breast tissues and the relationship between the expression and breast carcinoma.Method:Immunohistochemical staining SP method was used to detect the expression of p63 protein,p73 protein in 5 cases of normal mammary(controls),5 cases of hyperplasia mammary(hyperplasia),15 cases of carcinoma in situ(situ) and 60 cases of invasive breast carcinoma(invasive).Results:There was a continuous layer of myoepithelial cells in the benign lesions of breast,they were reactive for p63.Myoepithelial layer was interruptedly positive for p63 in carcinoma in situ but mostly or totally absent in early-invasive and invasive breast carcinoma.The expressions rates of p73 in the tissue of controls,hyperplasia,situ,and invasive were 0%(0/5),20.0%(1/5),33.3%(5/15)and 66.7%(40/60) respectively,and there were statistical significance compared to each other.With the increase of histological grade,clinic stage and lymphatic metastasis,the expression rate of p73 was increased.No correlation was found between p63 and p73 expression.Conclusion:p63 and p73 may play important roles in oncogenesis and progression of breast carcinoma.Detection of p63 and p73 protein may have potential effects on early diagnosis,direct treatment and prognosis of breast carcinoma.
