利用毛细管电泳-单链构象多态性快速鉴定血液感染病原菌的研究

摘    要:目的研究毛细管-单链构象多态性(capillary electrophoresis-single strand conformation polymorphism,CE-SSCP)分析技术鉴定血液感染病原菌的可行性,并建立一种快速检测病原菌的新方法。方法选取7种常见血液感染病原菌,在其16S rRNA的保守区设计通用引物,对16S rRNA基因进行扩增,然后将PCR产物进行CE-SSCP分析,依靠CE-SSCP图谱来鉴定病原菌。结果通用引物能够扩增出7种病原菌;CE-SSCP分析后每种菌具有不同出峰时间:大肠杆菌(3999);金黄色葡萄球菌(4082);铜绿假单胞菌(3958);表皮葡萄球菌(4063);肺炎链球菌(4055);肺炎克雷伯菌(3980);粪肠球菌(4166)。结论采用毛细管电泳-单链构象多态性分析技术可简便、快速地鉴定常见血液感染病原菌。

Abstract:Objective To investigate the feasibility of identifying pathogens in blood infection by capillary electrophoresis-based single strand conformation polymorphism (CE-SSCP), and to establish a new method for rapid identification of pathogens.
