高效液相色谱法测定直肠癌术中肠腔化疗血液和盆腔液及组织5-Fu浓度

目的:测定直肠癌术中肠腔化疗血液、盆腔液和组织5-Fu浓度。方法:20例直肠癌患者术中行肠腔化疗,注药后不同时相采集门静脉血、周围静脉血、盆腔液和癌旁组织,用高效液相色谱法测定5-Fu浓度。结果:门静脉血、盆腔液和组织2min即测得5-Fu,此后,盆腔液和组织浓度降低,门静脉血药浓度上升,60min达到峰浓度,周围静脉血未测得5-Fu。结论:直肠癌术中肠腔化疗可使5-Fu迅速分布于盆腔液、组织和门静脉血,但组织和盆腔液峰浓度低且下降迅速,对预防术后局部复发和腹腔转移作用有限,含5-Fu的门静脉血对抑制肝脏转移有一定意义。

Study on HPLC assay of 5-Fu concentration in patient's serum and peritoneal and tissue after intra-rectal chemotherapy of rectal tumor

Objective:To determine 5-Fu concentration in serum,peritoneal fluid and tissue by high performance liquid chromatography(HPLC) after intra-rectal chemotherapy of rectal tumor.Methods:20 cases with rectal tumor were underwent the treatment of intra-rectal chemotherapy.Portal veins,peripheral circulation blood,peritoneal fluid and tissues around tumor were sampled to analyze the 5-Fu content tested by our established HPLC method.Results:The portal veins,peritoneal fluid and tissues around tumor were tested 5-Fu on 2 minutes.Then the concentration of peritoneal fluid and tissues decreased,and the concentration of portal vein upgraded,reach maximum concentration on 60 minutes.5-Fu was not found in peripheral circulation blood.Conclusion:Intra-rectal chemotherapy of rectal tumor enable 5-Fu to be distributed to portal vein,peritoneal fluid and tissue quickly,but the maximum concentration of peritoneal fluid and tissues were low and dropped quickly,so had limited action to preventing post-operative metastasis to the cavity.Portal vein with 5-Fu has significance in preventing metastasis to liver.
