亚硒酸钠对U—937细胞增殖,分化及双链RNA含量的影响

摘    要:采用流式细胞仪检测等方法,初步研究亚硒酸钠对U-937细胞的抑制增殖作用和诱导分化作用及其对U-937细胞双链RNA的影响,结果提示:亚硒酸钠可抑制U-937细胞增殖,其作用随剂量增加和时间延长而增强。可使细胞用期发生改变,表现为G_0/G_1期细胞比例增加,S期细胞减少,G_2/M期细胞减少。可增强细胞非特异性酯酶和酸性磷酸酶活性,同时伴有单核细胞样形态变化,提示亚硒酸钠可能有诱导U-937细胞分化的作用。亚硒酸钠尚可明显降低U-937细胞双链RNA过量百分率。

Abstract:In the prescnt article, the effects of sodium selenite on the proliferation, differentiation and double-strandcd RNA of the human hitiocytic lymphoma cell line U-937 were reported. Thc results showed that the proliferation of U-937 could bc inhibited by sclenite in a dosc and time-dcpcndant manner, and the cell cyele changed. The biochemical analysis of the selenite treated cells demonstratcd that selenite raised the activi- ties of nonspecifie esterase and acidic phosphotase of thc cell line, which coincided with a monocytoid change in morphology. This suggested that sclenite can probably induce differentiation of U-937 to some extent. Flow cytometrie analysis of double-stranded RNA content distribution of U-937 showed that the cell line had a high percentage of double-stranded RNA excess, and the treatment with selenite resulted in a decrease of the percentage from 87% to 67%.
