摘    要:目的:对高原红细胞增多症(HAPC)骨髓前红系祖细胞(BFU—E)进行动态观察,以探讨其发病机理。方法:应用造血干细胞体外培养技术,取其体系内含单个核细胞2×10~5/ml置于培养液中,在5%CO_2、37℃及饱和湿度条件下培养,以观察其细胞生长情况。结果:红系祖细胞接种及增殖延滞时间为24h后,可见双细胞及少数4~6个细胞组成的细胞团。第4天可见10个左右细胞的细胞团,第6~7天可见较多CFU—E集落,第9天CFU—E集落数量明显减少,第10天可见较大或多中心的集落。呈桔黄色或颜色逐渐加深,以50~100个细胞的爆式集落为多见,联苯胺染色阳性,呈棕褐色,第14天可见较多较大的爆式集落含100~1000个左右细胞,培养至21d,BFU—E集落多数退化或消失。结论:观察中发现HAPC骨髓BFU—E集落产率与正常对照比较有显著差异(P<0.01),集落类型以多中心型,分散型为主。提示,HAPC骨髓BFU—E具有很强的增殖、分化能力,其机理需要进一步探讨。

Abstract:Objective: To study the mechanism of high altitude polycythemia(HAPC). Method: To observe dynamically the burst forming unit - erythroid(BFU - E) of HAPC patients in vitro. Result: The proliferative delay time was 24h behind inoculation of erythrogones. At this time, some double cells and a few cell balls formed by 4 to 6 cells were observed. On the 6th or 7th day, an number of colony forming unit - erythroid(CFU- E) were found and their number reduced obviously behind 9th day. On the 10th day, BFU-E was found. When having been cultured for 21 days, BFU - E degraded or disappeared. There were 3 colony types, i. e. thickening, multiple center and dispersion. The forming rate of BFU -E in HAPC group was higher than that of control group (P<0. 01). Conclusion: It is suggested that the proliferation and differentiation of BFU-E of HAPC patients increase obviously.
