水芹总酚酸对人肝癌2.2.15细胞周期的影响

摘    要:目的研究水芹总酚酸对人肝癌2.2.15细胞增殖抑制及细胞周期的影响。方法用不同浓度的水芹总酚酸作用于体外培养的2.2.15细胞,采用MTT法检测水芹总酚酸对2.2.15细胞增殖抑制能力;倒置显微镜观察2.2.15细胞的形态学变化;流式细胞术检测水芹总酚酸对2.2.15细胞周期的影响。结果水芹总酚酸对2.2.15细胞增殖具有明显的抑制作用,且具有剂量依赖性;流式细胞术检测表明水芹总酚酸作用于2.2.15细胞后,可将细胞阻滞在S期。结论水芹总酚酸对人肝癌2.2.15细胞具有明显的抑制增殖作用,可能与阻滞细胞周期有关。

Abstract:Objective To investigate the effect of total phenolics acid extracted from Oenanthe Javanica on cell growth and cell cycle in human hepatoma cell 2.2.15 in vitro.Methods The human cell 2.2.15 was cultured and treated by total phenolics acid extracted from Oenanthe Javanica at different concentrations in vitro.MTT assay was used to test dynamically the inhibitive effect on cell growth;morphologic changes were observed using the fluorescent microscope,and the modulation of cell cycle was measured by flow cytometry.Results Total phenolics acid extracted from oenanthe javanica had inhibitive effect on growth of 2.2.15 cells in a dose-dependent manner.Flow cytometry showed that total phenolics acid extracted from Oenanthe Javanica could block cell growth at S phase.Conclusion Total phenolics acid extracted from Oenanthe Javanica inhibits 2.2.15 cell proliferation,which is associated with the block of cell cycle.
