RP-HPLC法测定人血浆中头孢他美浓度

目的:建立以RP-HPLC法测定人血浆中头孢他美浓度的方法。方法:色谱柱为Hypersil C18,流动相为0.04%高氯酸溶液-乙腈(83∶17),柱温为40℃,流速为1.0mL.min-1,检测波长为265nm。结果:头孢他美检测浓度在0.125~8.0μg.mL-1范围内与峰面积线性关系良好(r=0.999 8),最低检出浓度为0.125μg.mL-1,平均回收率为100.6%(RSD=1.2%);日内RSD≤5.3%(n=5),日间RSD≤4.0%(n=5)。结论:本方法简便、灵敏、准确、重现性好,可为头孢他美酯胶囊药动学和生物利用度研究提供参考。

Determination of the Concentration of Cefetamet in Human Plasma by RP - HPLC

OBJECTIVE: To establish a RP-HPLC method for the determination of cefetamet in human plasma.METHODS: Hypersil C18 was used as column,and 0.04% perchloric acid-acetonitrile(83∶ 17) was taken as mobile phase.The column temperature was set at 40℃;The flow rate was 1.0mL· min-1 and the detection wavelength was 265nm.RESULTS: The calibration curve showed good linearity in a concentration range of 0.125～ 8.0μ g· mL-1(r=0.999 8),the minimum detectable concentration of cefetamet in plasma was 0.125μ g· mL-1,The average recovery rate was 100.6%(RSD=1.2%).The within-day RSD was less than 5.3%(n=5) and the inter-day RSD was less than 4.0%(n=5).CONCLUSION: The method is simple,sensitive,accurate and reproducibily and can be applied to the pharmacokinetic and bioavailability study of cefetamet pivoxil.
