共聚焦显微镜观察人羊膜匀浆提取液对大鼠碱烧伤后角膜超微结构的影响

摘    要:目的:通过角膜共聚焦显微镜活体动态观察人羊膜匀浆提取液对大鼠角膜碱烧伤后角膜超微结构的影响,探讨羊膜匀浆提取液应用于角膜碱烧伤后的临床价值及作用机理。方法 SD大鼠40只(40只眼),通过1 mol/L的氢氧化钠( NaOH)溶液建立右眼角膜碱烧伤模型。随机分为实验组和对照组,实验组行人羊膜匀浆提取液( AM)点眼,每日4次,对照组用PBS液点眼,每日4次。碱烧伤后行共聚焦显微镜动态观察大鼠角膜各层细胞数量以及形态的变化。结果实验组角膜上皮层炎症细胞形成晚,数量较少;基质层纤维结构排列较为规则,肌成纤维细胞的增殖和激活较少、角膜新生血管数量少,管径细。结论新鲜人羊膜匀浆提取液可以有效促进大鼠碱烧伤后角膜上皮细胞的增殖,抑制炎症细胞浸润,减少角膜新生血管的形成,抑制成纤维细胞的激活和增殖,减少角膜瘢痕的形成,有利于碱烧伤后大鼠角膜的透明愈合。

Abstract:Objective To observe ultrastructural changes in alkali burnt rat eyes treated with amniotic membrane ( AM) suspension , and to study the effective methods for eye alkaline burn treatment .Methods Forty eyes from 40 Spra-gue-Dawley healthy male rats were randomly assigned into control groups (20 eyes) or experiment groups (20 eyes).The animal model of alkali burn was set up with 1 mol/L NaOH.All eyes in the control groups received PBS four times a day , while in experiment group eyes were treated with amniotic suspension four with the same schedule .Tissue ultrastructural changes were analyzed with corneal confocal microscope .Results The CNV in experiment group emerged later and grew slower than that in control group .The area of CNV was significantly different between control group and experiment group at each time point .Inflammatory cells within cornea were significantly more in control group than in experiment group .The number of fibroblast cells of homogenate amnion was less in the experiment group .At 28th day, cellular density of endothe-lial cells was larger in experiment group .Conclusions AM suspension inhibits inflammatory cells , delays corneal neovas-cularization , restores corneal stromal structure , reduces the number of corneal stromal collagen fibers , and reduces the cor-neal scar .
