| 摘 要： | 目的探讨兔角膜缘干细胞培养方法，并获得兔角膜缘干细胞。方法采用组织块培养法，对兔角膜缘干细胞行体外培养；对原代培养第7d的细胞进行p63、AE5免疫组织化学鉴定。结果原代培养的细胞通常在7d左右即可达到80％～90％的融合，细胞呈椭圆形或多边形，核质比例大，表现出较强的增生能力。结论采用组织块培养法对兔角膜缘组织进行体外培养，可获得较纯、未分化、有增生能力的角膜缘干细胞。 |

| Abstract: | Objective To investigate a cultural method for rabbit corneal limbal stem cells in vitro and to obtain rabbit corneal limbal stem cells. Methods Rabbit corneal limbal stem cells were primarily cultured in vitro by using tissue block method till day 7, and then they were identified by immunohistochemical measurement of p63 and AE5. Results The primarily cultured cells usually achieved the fusion by 80% - 90% about 7 days, and they were oval or polygonal with large nuclear - cytoplasmic ratio demonstrating a strong proliferative capacity. Conclusion Pure, undifferentiated rabbit limbal stem cells with proliferative capacity can be obtained by using tissue block method. |
