微波对固体培养基灭菌效果的观察

摘    要:目的研究家用微波炉对农业用绿僵菌固体培养基灭菌效果。方法采用载体杀菌试验进行了实验室观察。结果随着负载量的增大,杀菌速度降低。负载量为200 g以下时,微波处理3 m in,全部无菌生长。负载量为250 g时,微波照射4 m in,存活菌数仍达100 cfu/g。试验证明,随着微波处理时间的延长,灭菌效果增强。以100 g固体培养基加60 g水的比例经微波处理效果比较好,灭菌处理3 m in均能达到灭菌目的。结论微波对绿僵菌固体培养基灭菌最佳工艺为:100 g的固体培养基加60 g水,浸润3 h,在800 W的微波功率处理3 m in,可达到灭菌效果。

Abstract:Objective In order to study the efficacy of household microwave oven in sterilizing agricultural Metarhizium anisopliae solid culture medium. Methods Carrier germicidal test was used to carry out laboratory observation. Results With increasing load,the germicidal speed decreased.At the load below 200 g,microwave treatment for 3 min resulted in no bacteria growth.At the load of 250 g,after microwave irradiation for 4 min,the surviving bacterial count was still 100 cfu/g.The test demonstrated that the sterilizing efficacy was enhanced with prolonging microwave irradiation time.The efficacy of microwave treatment was relatively good when 60 g of water was added to 100 g of the solid culture medium and sterilization could be achieved after sterilizing treatment for 3 min. Conclusion The best technology of microwave sterilization of Metarhizium anisopliae solid culture medium was adding water 60 g to solid culture medium 100 g,infiltration for 3 h and 800 W microwave treatment for 3 min.Sterilization can be achieved by this technology.
