NDRG1与Hpa在肝癌中的表达及与肝癌浸润转移的关系

目的探讨分化相关基因NDRG1和乙酰肝素酶(Hpa)在肝癌细胞中的表达及其与肝癌浸润、转移之间的关系。方法采用免疫组织化学方法检测46例肝癌、癌旁及正常肝组织中的表达,并分析二者表达情况与肝癌临床病理特征的关系,以及二者在肝癌组织中表达的相关性。结果 NDRG1和Hpa在正常肝组织及癌旁组织中弱表达或不表达,在肝癌组织中高表达。NDRG1蛋白表达水平与肿瘤分化程度和肿瘤大小无关,而与门静脉有无癌栓,肝内或淋巴结转移相关(P<0.05)。HPA蛋白表达水平与有无癌栓无明显相关性(P>.05),与肝癌组织分化程度,肿瘤大小,肝内或淋巴转移相关(P<0.05)。肝癌组织中NDRG1和Hpa的表达呈正相关(r=0.617,P<0.01)。结论 NDRG1和Hpa可能作为潜在癌基因在肝癌进展过程中起一定作用,联合检测NDRG1和Hpa蛋白的表达有助于判断肝癌生物学行为。

Expressions of NDRG1and HPA in hepatic carcinoma and their correlation with the infiltration and metastasis of hepatic carcinoma

Objective To investigate the expressions of N-myc downstream-regulated gene 1(NDRG1) and heparanases(HPA) in hepatocellular carcinoma(HCC) and to explore their correlations with the infiltration and metastasis of HCC.Methods The expressions of NDRG1and HPA in 46 cases of HCC,tumor-adjacent tissues,and normal liver tissues were detected by immunohistochemistry.The correlation between the expressions and clinicopathological features of HCC was analyzed,so was the correlation of expression between the two factors in hepatic carcinoma tissue.Results NDRG1and HPA were not expressed or little expressed in tumor-adjacent tissues and normal liver tissues,however,which were over-expressed in HCC tissues(P<0.05).The expression levels of NDRG1 in HCC were not correlated with tumor differentiation degree and tumor’s size,however,which were correlated with cancer embolus in portal vein and intrahepatic or lymphatic metastasis of HCC(P<0.05).The expression levels of HPA in HCC were not correlated with cancer embolus in portal vein(P>0.05),however,which were correlated with the differentiation degree of tumor tissues,tumor’s size,intrahepatic or lymphatic metastasis(P<0.05).The expression of NDRG1in hepatic carcinoma tissues was positively with that of HPA(r=0.617,P<0.01).Conclusion NDRG1and HPA as potential oncogenes may play a certain role in the progression of hepatic carcinoma,and combined detection of the expressions of NDRG1and HPA protein is helpful to evaluating the biological behaviors of HCC.
