凹叶厚朴细胞悬浮培养的研究

摘    要:目的研究凹叶厚朴MagnoliaofficinalisRehd.etils.Subsp.biloba(Rehd.etwils.)Law的细胞悬浮培养。方法以凹叶厚朴茎段诱导的愈伤组织为材料,应用L9(34)正交设计研究植物生长调节剂对凹叶厚朴悬浮细胞生长的影响,通过单因素试验,考察初始接种量、蔗糖浓度、pH值对凹叶厚朴悬浮细胞生长的影响。结果植物生长调节剂6-BA是影响凹叶厚朴悬浮细胞生长的主要因素;最佳初始接种量为8ml;最佳蔗糖浓度为30g/L;最佳pH值为6.5时。结论初步建立了适合凹叶厚朴悬浮细胞培养的体系。

Abstract:Objective To Study on the Cell Suspension Cultare ofM. officinalis Rehd. et Wils. Subsp. biloba (Rehd. et wils.) Law. Methods The callus ofM. officinalis Rehd. et Wils. Subsp. Biloba (Rehd. et wils. ) Law was selected as material, by L9 (34) orthogonal design, to study effects of plant growth regulators in file M. officinalis Rehd. et Wils. Subsp. Biloba (Rehd. et wils.) Law cell suspension culture; by single factor analysis, to study effects of the initial inoculation, sugar content, the pH value in file M. officinalis Rehd. et Wils. Subsp. Biloba (Rehd. et wils.) Law cell suspension culture. Results The results showed that 6-BA was the most important effect fact in the four plant growth regulators, and that the most initial inoculation was 8ml, and that the most sugar content was 30 g / L, and that the most the pH value was 6.5. Conclusion An M.officinalis Rehd. et Wils. Subsp. Biloba (Rehd. et wils.) Law suspension cell culture system was preliminarily established.
