干姜、大黄和丹参水提物对MPP^+诱导的SH-SY5Y细胞凋亡的药性学对比研究

摘    要:目的研究干姜、大黄和丹参水提物对四氢吡啶离子(MPP+)诱导SH-SY5Y细胞凋亡的影响。方法体外培养SH-SY5Y细胞,建立MPP+致SH-SY5Y细胞凋亡的模型,同时给予干姜、大黄和丹参水提物干预。应用MTT比色法检测细胞活力,流式细胞仪检测细胞凋亡率。结果 0.5mmol.L-1MPP+作用于SH-SY5Y细胞60h,MTT值降低到(30.43±2.69)%(P〈0.01vs.control),Hoechst33258染色可见明显的颗粒状和固缩状荧光,凋亡率增加到(40.52±1.68)%(P〈0.01vs.control),而干姜水提物能明显改善以上的凋亡特性,显著升高其MTT值,降低凋亡率(P〈0.05vs.modelgroup),大黄和丹参水提物则对MPP+诱导的SH-SY5Y细胞凋亡无保护作用。结论干姜对MPP+诱导的SH-SY5Y细胞凋亡有显著保护作用,而大黄和丹参却无此保护作用,中药药性不同,其抗凋亡作用亦不同。

Comparative Study on Four Natures of the Protective Effects of Water Extracts of Zingiber officinale,Rheum Palmatum and Salvia Miltiorrhiza on SH-SY5Y Cells Apoptosis Induced by MPP+

Abstract:Objective To investigate the protective effects of water extracts of Zingiber officinale, Rheum palmatum and Salvia miltiorrhiza on SH-SY5Y cells apoptosis induced by MPP+ in culture. Methods SH-SY5Y cells were cultured in vitro and MPP+ induced SH-SY5Y cells apoptotic model was established and treated with Water extracts of Z. officinale, R. palmatum and S. miltiorrhiza. The cell Viability was measured by MTT test, the morphology of SH-SY5Y cells was observed by Hoechst33258 staining and the percentage of apoptosis by FCM. Results The cell activity decreased remarkably to (30.43±2.69) % (P〈0.01 vs. control) induced by 0.5 mmol/L MPP+ for 60 h and were positively stained with fluorochrome Hoechst 33258, presenting the percentage of apoptosis as (40.52 ± 1.68) % (P 〈 0.01 vs. control), However, water extracts of Z. officinale obviously improved MTT value, reduced the percentage of apoptosis (P 〈 0.05 vs. model group), and water extracts of R. palmatum and S. miltiorrhiza, almost had no effect on the apoptosis of SH-SY5Y cells. Conclusion Z. officinale has markedly protective effects on SH-SY5Y cells apoptosis induced by MPP+, but R. palmatum and S. miltiorrhiza, show no protective effects, as the different natures of TCM, the anti-aooototic effects of TCM are different neither.
