摘    要:目的:探讨肿瘤坏死因子-α(TNF-α)在铜绿假单胞菌肺部感染发病中的作用。方法:90只小鼠随机分成对照组、模型组与抗体组,每组30只。通过气管插管,在气管内注入铜绿假单胞菌建立小鼠肺部感染模型,抗体组手术前6h静脉内注入0.2mL/kgTNF-α抗体,对照组注入相同剂量的PBS液,分别在0、3、6、12、24h不同时间点收集支气管肺泡灌洗液,用ELISA法检测各组不同时间点支气管肺泡灌洗液中TNF-α的含量,并观察小鼠1周后的死亡率。结果:模型组和抗体组支气管肺泡灌洗液中TNF-α含量在3h时已开始升高,6h达到高峰,12、24h逐渐降低,模型组明显高于抗体组(P〈0.05—0.01),而两组均显著高于对照组(P〈0.01)。1周后模型组小鼠死亡率(73.3%)明显高于抗体组(43.3%)(P〈0.01),两组死亡率均显著高于对照组(3.3%)(P〈0.01)。结论:TNF-α在铜绿假单胞菌肺炎发病过程中起着非常重要作用,抑制其过度释放可减缓病情。

Abstract:Objective:To investigate the role of tumor necrosis factor-α(TNF-α)in the course of pseudomonas aeruginosa pneumonia. Methods:90 mice were randomly divided into three groups:model group,antibody group and control group. Pseudomonas aeruginosa strain was instilled into the trachea of the mice through tracheal cannula to induce pneumonia in each group. However,6 hours before the instillation,TNF-α antibody and PBS liquid were injected (0.2 mL/kg,iv) to the antibody group and the control group,separately. Bronchoalveolar lavage fluid (BALF) were harvested at 0 h,3 h,6 h,12 h,24 h after pneumonia was induced,and measured for the TNF-αconcentration with enzyme-linked immunosorbent assay (ELISA) in each group. Results:In both model group and the antibody group,TNF-αlevel of BALF started to increase gradually 3h after pneumonia onset,reached the peak at 6h,decreased after 12h,and were significant higher than that of the control group (P<0.01). Between the model group and the antibody group,TNF-α level of the former was significant higher (P<0.05~0.01). After a week,the mortality of the model group(73.3%) was significant higher than that of the antibody group(73.3%) and the control group(3.3%),among the latter two,antibody group showed higher mortality(P<0.01). Conclusion:Suppression excessive TNF-α release can relieve pneumonia,as TNF-αhas an important role in the course of pseudomonas aeruginosa pneumonia.
