复方夏决提取物对肾性高血压模型大鼠心脏血流动力学的影响

摘    要: 目的观察复方夏决提取物对麻醉肾性高血压大鼠心脏血流动力学的影响。方法SD大鼠采用肾动脉狭窄法造模,6周后制成肾性高血压模型(RHR)。肾性高血压大鼠随机分成5组:生理盐水组、阳性对照组(卡托普利15 mg·kg-1)、复方夏决提取物3个剂量组(3.6,5.4,7.2 g·kg-1),每组10只。麻醉状态下分离大鼠颈总动脉和股动脉,并进行插管。采用十二指肠给药,分别观察给药后0,15,30,45,60,90,120 min,心脏血流动力学各相应指标如收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)、平均脉压差(MPP)、左心室收缩压峰值(LVSP)、左室舒张末期压(LVEDP)、左室等容期压力最大变化速率(t±dp/dtmax)、左室开始收缩至左室内压上升速率峰值时间(t-dp/dtmax)、心率(HR)的变化。结果复方夏决提取物不同剂量均能使麻醉大鼠的SBP、DBP、MAP、LVSP、LVEDP、±dp/dtmax不同程度地降低(P<0.05),t-dp/dtmax延长(P<0.05),但对MPP、HR几乎无影响。结论复方夏决提取物降压作用与改善心脏舒张功能及抑制心肌收缩有关。

Abstract:OBJECTIVE To observe the effect of extract from compound Xiajue on cardiac hemodynamics in anesthetized renovascular hypertensive rats.METHODS SD rats were turned into renovascular hypertensive rats(RHR) by chronic partial occlusion of left renal artery after 6 weeks.RHR were randomly divided into 5 groups: negative control group(NS),positive control group(captopril,15 mg·kg-1) and 3 groups treated with the extract from compound Xiajue(FJM 3.6,5.4,7.2 g·kg-1),10 rats each group.In anesthetic conditions,right-sidedness common carotid artery and femoral artery of RHR were separated to intubate.Cardiac hemodynamics indexes such as systolic blood pressure(SBP),diastolic blood pressure(DBP),mean arterial pressure(MAP),mean pulse pressure(MPP),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),maximal rising and falling rate of ventricular pressure(±dp/dtmax),t-dp/dtmax,heart rate(HR) were determined at 0,15,30,45,60,90,120 min after duodenal administration. RESULTS Three FJM groups(3.6,5.4,7.2 g·kg-1) decreased the anesthetic RHR's cardiac hemodynamics indexes such as SBP,DBP,MAP,LVSP,LVEDP,±dp/dtmax(P<0.05,or P<0.01),prolonged t-dp/dtmax(P<0.05),hardly had influence on MPP and HR.CONCLUSION The anti-hypertesion action mechanism of FJM may be related to improve cardiac diastolic function and inhibiting the myocardiac contraction.
