基于PI3K/Akt信号转导通路的通脉醒脑滴丸治疗脑缺血的机制研究

摘    要:目的:探讨中药复方制剂通脉醒脑滴丸防治脑缺血卒中的作用机理。方法:选用线栓阻断大鼠大脑中动脉法复制脑缺血再灌注模型,采用荧光定量PCR方法测定脑组织PI3K、Akt和PTEN的表达,并通过免疫组化方法测定脑组织HIF-1a和VEGF的表达,考察通脉醒脑滴丸对PI3K/Akt信号通路的作用。结果:通脉醒脑滴丸能上调脑组织PI3K、Akt、HIF-1a以及VEGF的表达,下调PTEN的表达。结论:研究初步证实通脉醒脑滴丸对脑缺血损伤具有保护作用,其机制可能与介导PI3K/Akt细胞存活通路的激活或上调有关。

Abstract:Objective This study aims to explore the mechanism of Tongmai Xingnao Dripping Pill treating ischemia cerebral vascular disease (ICVD). Methods The model of middle cerebral artery occlusion (MCAO) by suture embolism in rats were used. We mensurated the expressions of PI3K, Akt and PTEN in brain by RT-PCR and the expressions of HIF-1 a and VEGF in brain by immunohistochemistry to observe the effection of the Tongmai Xingnao Dirpping pill on the pathway of PI3K/Akt. Results Tongmai Xingnao Dripping Pill can up-regulate the expressions of PI3K, Akt, HIF-1 a and VEGF and down-regulate the expression of PTEN. Conclusion The results can elementarily confirm that Tongmai Xingnao Dripping Pill has protective effect of the damages of ICVD and it is probably related to the up-regulation of PI3K/Akt pathway.
