姜黄素对溃疡性结肠炎脂质过氧化损伤大鼠的保护作用

摘    要:目的探讨姜黄素对大鼠溃疡性结肠炎(UC)脂质过氧化损伤的保护作用。方法采用三硝基苯磺酸(TNBS)灌肠制备大鼠UC模型,将动物随机分为正常对照组,三硝基苯磺酸组,三硝基苯磺酸+生理盐水组,三硝基苯磺酸+姜黄素组4组。观察各组大鼠肠道大体形态和组织学改变,分别测定血清和肠组织中超氧化物岐化酶(SOD)活性和脂质过氧化产物丙二醛(MDA)含量。结果结果显示UC大鼠SOD活性显著低于正常对照组(P〈0.01),MDA含量显著高于正常对照组(P〈0.01);姜黄素治疗组的SOD活性显著高于UC组(P〈0.01),MDA含量显著低于UC组(P〈0.01)。病理学显示姜黄素治疗组有明显好转。结论姜黄素通过抑制脂质过氧化反应对UC有保护作用,抗氧化应激可能是其作用机制之一。

Abstract:Objective To observe the effects of Curcumin(Cur) on lipid peroxidation in ulcerative colitis of rats.Methods Rat ulcerative colitis model was induced by trinitrobenzenesulfacid(TNBS).The rats randomly divided into 4 groups:normal,TNBS,TNBS+NS and TNBS+Cur group.The macroscopical and hisological changes of the colon were evalucated.The superaoxide dismutase(SOD) activity and the malondialdehyde(MDA) content in the colonic mucosa and serum were determined,and the indexes were compared with those in normal group and control group with ulcerative colitis.Results It showed that SOD activity in the UC group was much lower than that in the normal group(P<0.01)but MDA content much higher(P<0.01);and the SOD activity in the Cur group was much higher than that in the UC group (P<0.01) but the MDA content much lower(P<0.01).The pathological examination showed that local changes of UC were looked better after Cur treatment.Conclusion Curcumin has protective effects on ulcerative colitis in rats by inhibiting lipid peroxidation,it may be related to resisting oxidative stress.
