补肾活血方通过p38 MAPK信号通路影响人成骨细胞功能的机制

摘    要:目的:观察补肾活血方含药血清对人成骨细胞功能及其对p38 MAPK信号转导通路的活性影响,探讨补肾活血方防治骨质疏松症的作用机制。方法:将人成骨细胞株分为Control组、补肾活血方含药血清组(Z组)、含药血清+p38抑制剂SB203580组(Z+SB组),采用四甲基偶氮唑盐比色法(MTT)检测各组成骨细胞的增殖情况,通过PNPP法分析检测成骨细胞碱性磷酸酶(ALP)的活性,采用免疫印迹(Western blot)法检测和磷酸化p38(p-p38)的表达水平。结果:补肾活血方含药血清能刺激成骨细胞的增殖,使分化增强,与Control组比较有显著性差异(P〈0.01);阻断p38 MAPK信号转导通路后,与Z组比较,对成骨细胞的增殖无明显抑制(P〉0.05),而对成骨细胞的分化功能有显著抑制作用(P〈0.01);Western blot结果显示,补肾活血方含药血清能刺激p-p38表达,阻断p38 MAPK信号转导通路后,p-p38含量明显减少(P〈0.01)。结论:补肾活血方可能通过p38 MAPK通路调控促进成骨细胞分化的功能,起到防治骨质疏松症的作用。

Abstract:Objective:To observe the effect of Bushenhuoxue decoction medicated serum on human osteoblasts function and activity of p38 MAPK signal transduction pathway, and investigate the mechanism of the preventing and treating osteoporosis by Bushenhuoxue decoction. Methods:Divid-ed human osteoblasts strains into Control group, Bushenhuoxue decoction medicated serum group (group Z), medicated serum+p38 inhibitor SB203580 group (group Z+SB), used MTT to test the proliferation of osteoblasts in each group, analyzed the activity of osteoblasts' ALP by PNPP, and tested the expression level of p-p38 by Western blot. Results:Bushenhuoxue decoction medicated serum could stimulate the proliferation of os-teoblasts and make its differentiation being enhanced, there were significant differences that compared with Control group (P〈0.01);after blocking p38 MAPK signal transduction pathway, compared with group Z, the proliferation of osteoblasts had no significantly restrained (P〉0.05), but there was significant inhibition effect on the differentiation of osteoblasts (P〈0.01);the Western blot result showed that Bushenhuoxue decoction medicat-ed serum could stimulate the expression of p-p38, and the content of p-p38 would be significantly decreased by blocking p38 MAPK signal transduc-tion pathway (P〈0.01). Conclusion:Bushenhuoxue decoction may promote the differentiation of osteoblasts with regulation of p38 NAPK pathway to prevent and cure osteoporosis.
