肾炎康复片对TGF-β_1诱导下肾小球系膜细胞增殖及细胞外基质合成的影响

摘    要:目的:观察肾炎康复片对转化生长因子-β1(transforming growth factor,TGF-β1)诱导下肾小球系膜细胞增殖及细胞外基质成分表达的影响。方法:体外培养正常大鼠肾小球系膜细胞,分为正常对照组,TGF-β1刺激组,TGF-β1加肾炎康复片低剂量组(100 mg·L^-1)、中剂量组(200 mg·L^-1)、高剂量组(500 mg·L^-1)。分别利用甲基噻唑基四唑法(metyl thiazolyl tetronzolium,MTT)观察肾炎康复片对TGF-β1诱导细胞活力的影响;双抗体夹心ABC-ELISA法检测细胞上清I型胶原(Col I)、纤维连接蛋白(FN)的含量。结果:TGF-β1可以显著增加大鼠肾脏系膜细胞活力,TGF-β1刺激组Col I、FN表达量较正常对照组明显升高(P〈0.05);TGF-β1加肾炎康复片各剂量组的Col I、FN均低于TGF-β1刺激组(P〈0.05)。结论:肾炎康复片能抑制TGF-β1诱导的细胞活力及肾小球细胞外基质(ECM)的合成。提示抑制TGF-β1刺激GMC分泌细胞外基质是肾炎康复片治疗慢性肾小球疾病的重要机制之一。

Abstract:Objective: To observe the influence of Shenyankangfu Tablet on glomerular mesangial cell proliferation and extracellular matrix synthesis induced by TGF-beta 1. Methods: In vitro cultured normal rat mesangial cells,divided into normal control group,TGF-beta1 stimulation group,TGF-beta 1 plus Shenyankangfu Tablet low dose group( 100 mg·L- 1),middle dose group( 200 mg·L- 1),high dose group( 500 mg· L- 1). Used methyl thiazolyl tetrazolium( MTT) respectively to observe the effect of Shenyankangfu Tablet on TGF-beta 1 induced cell viability; Double antibody sandwich ABC-ELISA to detect collagen type I( Col I),fibronectin( FN) content.Results: TGF-beta 1 can significantly increase the activity of glomerular mesangial cells of rats,in the TGF-beta1 stimulation group,the expressions level of Col I and FN were significantly higher than the normal control( P〈0. 05); Those of TGF-beta 1 and nephritis tablets in each dose group were lower than TGF-beta 1 stimulation group( P〈0. 05). Conclusion: Shenyankangfu Tablet can inhibit cell viability induced by TGF-beta 1 and glomerular extracellular matrix( ECM) synthesis,which suggest that inhibition of TGF-beta 1 stimulates GMC secretion of extracellular matrix is one of the important mechanisms of Shenyankangfu Tablet treatment of chronic glomerular disease.
