摘    要:目的:建立HPLC-ELSD法同时测定细柱五加果实中的竹节参苷Ⅳa和3β-(O-β-D-glucopyranuronosyl] oxy)olean-12-ene-28-olc acid含量的方法.方法:采用反相高效液相色谱-蒸发光散射检测法.色谱条件AlltimaTM-C18色谱柱(4.6mm× 250 mm,5μm),流动相乙腈(A)-0.5%乙酸水溶液(B),柱温35℃.ELSD检测器条件:漂移管温度106℃,载气氮气,流速2.9 L·min-1,放大系数1,撞击器关闭.结果:竹节参苷Ⅳa和3β-( O-β-D-glucopyranu ronosyl] oxy)-olean-12-ene-28-olc acid的进样量分别在0.535~ 8.560 μg(r=0.996 5)和0.515~8.240 μg(r=0.9987)呈良好线性关系;平均回收率(n=6)分别为98.39%,98.97%,RSD分别为1.50%,1.59%.结论:方法准确、灵敏,可用于测定细柱五加果实中竹节参苷Ⅳa和3β-( O-β-D-glucopyranu-ronosyl] oxy) -olean-12-ene-28-olc acid的含量.

Abstract:Objective:To establish an HPLC-ELSD method for the simultaneous determination of the content of chikusetsusaponin Ⅳa and 3β-(O-β-D-glucopyranuronosyl]oxy)-olean-12-ene-28-olc acid in the fruits of Acanthopanax gracilistylus.Method:The RP-HPLC-ELSD method was used.Chromatographic conditions:AlltimaTM-C18 chromatographic column(4.6 mm×250 mm,5 μm);acetonitrile(A)-0.5% acetic acid water(B) as mobile phase;column temperature at 35 ℃.ELSD detector conditions:drift tube temperature:106 ℃;carrier gas(nitrogen) flow rate:2.9 L · min-1;amplification factor:1;impactor:close.Result:The calibration curves showed good linear relationship in the range from 0.535 to 8.560 μg(r=0.996 5) for chikusetsusaponin Ⅳa and 0.515 to 8.240 μg(r=0.998 7) for 3β-(O-β-D-glucopyranuronosyl]oxy)-olean-12-ene-28-olc acid.The average recovery rates(n=6) were 98.39% and 98.97%,and RSDs were 1.50% and 1.59%.Conclusion:This method is accurate,sensitive and can be used to determine the content of chikusetsusaponin Ⅳa and 3β-(O-β-D-glucopyranuronosyl]oxy)-olean-12-ene-28-olc acid in the fruits of Acanthopanax gracilistylus.
