摘    要:目的:四逆散传统汤剂和配方颗粒是在临床中同时存在的两种用药形式,本研究拟对比研究四逆散传统汤剂和配方颗粒的疏肝解郁药效作用,为四逆散配方颗粒的临床使用提供实验依据.方法:昆明种小鼠随机分为空白组、应激模型组、阳性药组、四逆散传统汤剂低、高剂量(600,1200 mg·kg-1)组、四逆散配方颗粒汤剂低、高剂量(600,1200 mg·kg-1)组共7组,每组10只.小鼠连续灌胃给药1周,实验第6天时给予小鼠一次性拘束应激18h,测定血浆丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)水平.昆明种小鼠随机分为空白组、粉刺杆菌-脂多糖(Propionibacterium acne-LPS)致肝炎模型组、阳性药组、四逆散传统汤剂低、高剂量(600,1200 mg· kg-1)组、四逆散配方颗粒汤剂低、高剂量(600,1200 mg· kg-1)组共7组,每组10只.小鼠尾静脉注射20 mg·kg-1热灭活粉刺杆菌生理盐水溶液5d后,尾静脉注射5μg· kg-1脂多糖生理盐水溶液,5h测定血浆ALT,AST水平.结果:与正常对照组相比,应激模型组和P.acnes-LPS的血浆ALT均显著增高(43.4±2.0)U·L-1vs (12.2-2.8) U·L-1,P <0.05;(114.9±23.3) U·L-1vs (18.8±5.2) U·L-1,P <0.05,AST水平也显著增高.与模型组相比,四逆散传统饮片汤剂和配方颗粒均能显著降低应激负荷和粉刺杆菌-脂多糖升高的血浆ALT和AST水平,但是两药药效没有统计学差异.结论:四逆散传统饮片汤剂和配方颗粒均有较好的保肝作用.

Abstract:Objective:To compare the liver-protective effects between traditional decoction and Chinese medicinal dispensing granule of Sini San.Method:Liver injury models were induced by establishing restraint stress and Propionibacterium acnes-lipopolysaccharide(LPS) in mice.Kunming mice were divided into 7 groups,normal control,stress model,positive control,Sini San decoction(600,1 200 mg · kg-1) and Sini San dispensing granule(600,1 200 mg · kg-1).Mice were administered for consecutive 6 days and restrained for 18 h.Hepatic function was evaluated by assessing assessing alanine aminotransferase(ALT) and aspartate aminotransferase(AST) level in plasma.Kuming mice were divided into 7 groups,such as normal control,P.acnes-LPS model,positive control,Sini San decoction(600,1 200 mg · kg-1) and Sini San dispensing granule(600,1 200 mg · kg-1).Mice were injected with 20 mg · kg-1 P.acnes and 5 μg · kg-1 LPS to induce liver injury.Hepatic function was evaluated by ALT and AST level in plasma.Result:Compared with the model groups,the traditional decoction and Chinese dispensing granule of Sini San significantly reduced ALT and AST levels induced by restraint stress or P.acne-LPS.However,there were no significant difference between the two samples.Conclusion:The traditional decoction and Chinese medicinal dispensing granule of Sini San both are effective on improving the liver function in liver-injury mice model.
