阿魏酸镧配合物对H2 O2氧化损伤的血管内皮细胞的影响

摘    要:目的:探讨阿魏酸镧配合物( Lanthanum Ferulate,LF)对氧化损伤的血管内皮细胞的影响。方法体外培养内皮细胞,将细胞分为7组,即空白对照组、溶剂对照组( DMSO)、氧化损伤组( H2 O2)、氧化损伤加阿魏酸对照组、氧化损伤加阿魏酸镧配合物低、中、高浓度组( LF- L+ H2 O2、LF- M + H2 O2、LF- H + H2 O2)。阿魏酸及不同浓度阿魏酸镧配合物预培养内皮细胞24 h后,加入200μmol /L H2 O2继续培养12 h,用MTT法观察阿魏酸镧配合物对H2 O2损伤的内皮细胞活性的影响,检测各组细胞培养液内乳酸脱氢酶( LDH)、一氧化氮( NO)、丙二醛( MDA)含量。结果阿魏酸镧配合物呈剂量依赖性降低H2 O2对内皮细胞生长抑制率,降低MDA、LDH量,增加培养液中NO-2/NO-3含量,各指标差异有统计学意义(P〈0.05)。结论阿魏酸镧配合物可拮抗和修复过氧化氢诱导的血管内皮细胞的损伤,其作用可能与其抗氧化和促进NO释放有关。

Effects of lanthanum ferulate on endothelial cell injury induced by H2 O2

Abstract:Objective To study the effect of lanthanum ferulate( LF) on endothelial cell injuries induced by H2 O2 . Methods Endothelial cells were cultured in vitro and divided into seven groups: control,DMSO, H2 O2 ,ferulate+H2 O2 , LF-Low+H2 O2 , LF-Middle+H2 O2 and LF-High+H2 O2 . Endothelial cells were incu_bated in the absence or presence of various concentrations of lanthanum ferulate and ferulate for 24h, then cells were incubated with 200 μmol/L H2 O2 for 12h. Cell viability was tested by MTT. The amount of ma_londialdehyde ( MDA) , nitric oxide ( NO) , lactate dehydrogenase ( LDH) in serum, and cell viability were measured. Results Incubation endothelial cells with H2 O2 for 12h elicited the increase in the rate of apopto_sis, the release of LDH, and the decrease in the content of NO in the medium, which were inhibited by pre_treatment of lanthanum ferulate at concentrations of 5-125μmol/L in a concentration-dependent manner ( P〈0 . 05 ) . Conclusion lanthanum ferulate possesses a protective effect against endothelial cell in juries in_duced by H2 O2 , which may be related with anti-oxidation and increasing the concentration of NO.
