子宫内膜癌中CDK4及相关蛋白的表达情况及意义

目的通过检测CDK4、ki67和PTEN在子宫内膜癌组织中表达情况来探讨其意义。方法应用免疫组织化学染色S-P法检测60例子宫内膜癌、32例正常子宫内膜组织中三者的表达情况,计算阳性率并进行分析。结果子宫内膜癌组织中CDK4和ki67表达定位于细胞核,其阳性表达率分别为78.33%(47/60)、85.00%(51/60),明显高于正常子宫内膜组织25.00%(8/32)和12.50%(4/32),差异有统计学意义(P0.05)。PTEN蛋白在子宫内膜癌组织中缺失率为73.33%,高于正常子宫内膜组织(P0.05)。CDK4和ki67在不同组织学分级、不同临床病理分期以及是否有淋巴结转移的子宫内膜癌中的表达,差异均有统计学意义,(P均0.05);而在不同组织学亚型及不同肌层浸润程度中的表达,差异无统计学意义(P均0.05)。PTEN蛋白表达在肌层浸润深度、组织学亚型、淋巴结转移及手术-病理分期中差异无统计学意义(P0.05),而且PTEN表达在G1级肿瘤高于G2、G3级(P0.05),不同肿瘤组织学亚型无关,PTEN蛋白缺失率差异无统计学意义(P0.05),在子宫内膜癌组织中CDK4和ki67两者表达呈正相关(P0.05),CDK4与PTEN的表达无明显相关性。结论 CDK4和ki67在子宫内膜癌呈高表达和PTEN蛋白表达缺失,且与子宫内膜癌的发生、发展和转移具有一定的关系。

Expression and significance of CDK4 and PTEN protein in endometrial carcinoma.

Objective To determine the expression of CDK4 ki67 and PTEN in the endometrial carcinoma and the significance. Methods The expressions of CDK4 ki67 and PTEN in 60 endometrial cancer cases and 32 normal endometrial tissues were determined by immunohistochemistry S- P and the positive expression rates were analyzed. Results The expressions of CDK4 and ki67 were located with the cytoplasm and the endometrial. The positive expression rates were 78.33%（47/60）、85.00%（51/60） which were significantly higher than that in normal endometrial tissues in 25.00%（8/32）和12.50%（4/32）（P0.05）. Both of their expressions were related with myometrial invasion, lymph node metastasis, clinical phasing and pathological staging（P0.05）. Outcome of endometrial carcinoma tissue in PTEN protein expression for loss, higher than that in normal endometrial tissue（P0.05）, and the PTEN expression in G1 level was higher than that of G2, G3 tumor（P 0.05）,PTEN protein loss rate and tumor tissue types, and myometrial invasion, lymph node metastasis and pathological staging showed no significant relationship（P0.05）;In endometrial carcinoma, CDK4 and PTEN expression was significantly negative correlated,the expressions of CDK4 and ki67were positively correlated with each other. Conclusion CDK4 and ki67 were highly expressed and loss of PTEN protein in the endometrial cancinoma and their expressions were related with occurrence and development of endometrial carcinoma and the metastasis.
