结核患者血浆和胸水上清C1qC的表达水平的研究

目的利用酶联免疫吸附(ELISA)技术检测不同人群血浆中C1qC的蛋白表达水平以及结核病人胸水和血浆C1qC的蛋白表达水平。通过比较C1qC在不同人群和不同标本中的表达情况,评价其在结核诊断中的应用价值。方法利用ELISA技术分别对20例健康对照(HC)、20例结核潜伏感染者(LTBI)和27例肺结核病人(TB)血浆C1qC的表达水平;随访其中10例抗结核治疗的结核病人,检测其血浆中C1qC的表达,观察在抗结核治疗过程中(抗结核治疗前、抗治疗3个月和6个月)C1q C的表达水平;检测16例结核性胸膜炎病人配对的血浆和胸水上清中C1qC的表达水平,比较不同样本中C1qC的表达水平。结果 C1qC在结核健康对照、潜伏感染者、结核病人血浆中的表达水平分别为603.8±134.3、507.0±141.2、756.6±261.2(pg/mL)。肺结核病人C1qC的表达水平显著高于健康对照(P0.05)和潜伏感染者(P0.001);在10例结核病人中,C1qC在抗结核治疗前的表达水平为769.6±398.8(pg/mL),抗结核治疗3个月和6个月后C1q C表达分别为624.4±208.5和444.8±163.8(pg/mL),在抗结核治疗6个月后C1qC的表达水平显著地低于抗结核治疗前的水平(P0.05);C1qC在胸水上清中的表达水平显著高于血浆中的表达水平(P0.05)。结论 C1qC在结核患者的血浆中高表达,在抗结核治疗中逐渐下降,并且在结核感染部位表达显著增高。这些结果提示利用ELISA检测血浆和胸水上清中C1q C的表达可用于肺结核的辅助诊断。

Profiles of C1q C expression in the plasma and pleural fluid from tuberculosis patients

Object To detect and compare the expression of C1qC in the plasma and plasma pleural fluid from different populations and pulmonary tuberculosis patients by ELISA.Methods The level of C1qC in plasma from healthy controls（n=20）,latent tuberculosis infection（n=20） and patients with pulmonary tuberculosis（n=27） were detected by ELISA.Among of them,the C1qC expression in plasma from 10 patients with pulmonary tuberculosis before and 3 months and 6 months after anti-tuberculosis treatment was compared.At the same time,the level of C1qC in plasma and the paired pleural fluid from the patient with pulmonary tuberculosis（n=16） were detected.Results The level of C1qC in the plasma from healthy control,latent tuberculosis infection and patients with pulmonary tuberculosis were 603.8±134.3,507.0±141.2,756.6±261.2（pg/ml）.The level of C1qC in the plasma from tuberculosis patients was significantly higher than that from healthy control（P0.05） and latent tuberculosis infection（P0.001）.The level of C1qC in the plasma from tuberculosis patients before anti-tuberculosis therapy 3 months and 6 months after anti-tuberculosis therapy was 769.6±398.8,624.4±208.5,444.8±163.8（pg/ml）.Its level decreased 6 months after anti-tuberculosis therapy（P0.05）.And its expression in pleural fluid was significantly higher than in plasma（P0.05）.Conclusions Detection of C1qC level by ELISA could be useful for the diagnosis of pulmonary tuberculosis.
