右美托咪啶对大鼠肾缺血/再灌注肾功能的影响

摘    要:目的探讨右美托咪啶对大鼠肾缺血/再灌注后肾功能及磷酸化STAT3蛋白表达的影响。方法 SPF级SD大鼠30只,随机分为缺血再灌注组(I/R组)、假手术组(S组)和右美托咪啶预处理组(Dex组),每组10只。I/R组切除右肾,左肾缺血45min,再灌注60min,造肾I/R模型;Dex组于造模前泵注30min Dex;每组于I/R后6h取肾脏制作病理切片,HE染色观察;另取血样,分离血浆,多功能生化仪检测肌酐(Scr)和尿素氮(BUN)含量;ELISA检测肾损伤分子-1(Kim-1)、IL-1β、TNF-α及P-STAT3含量;Western-blot检测肾组织中P-STAT3表达的变化。结果 HE染色结果显示,与S组相比,I/R组肾脏损伤严重,炎症明显,而Dex干预组则明显减轻;肾功能及炎性因子检测结果显示,与S组比较,I/R组和Dex组Scr、BUN、Kim-1、IL-1β、TNF-α及P-STAT3含量均显著升高(P0.05),Dex组明显低于I/R组(P0.05)。Western-blot检测结果显示,I/R组肾脏中P-STAT3表达增高,Dex可降低P-STAT3表达水平。结论 Dex可以减轻由I/R引起的肾功能障碍,抑制炎性反应,降低磷酸化STAT3表达,对肾脏有一定的保护作用。

Abstract:Objective To explore the effect of Dexme detomidine on renal function and P-STAT3 expression in rats withrenal ischemial reperfusion. Methods SPF SD rats(n=30) were randomly divided into isocheimal reperfusion (I/R group) ,control group (S) and dexmedetomidine group ( Dex group). The right kidney of ischemia-reperfusion model was cut off and theleft one was ischemia for 45rain, then transfusing blood for 60 minutes. Dex group were reperfused group with Dex interventionfor 30 minutes. After IJR for a period of 6h, kidneys of rats were taken and pathological section were produced and observedwith HE staining; blood and plasma was separated, multi biochemical analyzer was used for detection of creatinine (Scr) andblood urea nitrogen (BUN) content. The kidney injury molecule -1 (Kim-1), IL-Iβ, TNF-a, P-STAT3 content in serum weredetected by ELISA. The expression of P-STAT3 was detected by Western-blot. Results Compared with S group, thedamage of kidneys if rats of UR group was serious, but that of Dex intervention group was significantly reduced. Compared withS group, Scr, BUN, Kim-1, IL-Iβ, TNF-ct and P-STAT3 content in the I/R group and Dex group were significantly increased(P 〈 0.05), but it was lower in Dex group than that in group I/R (P 〈 0.05).The content of P-STAT3 was obviously higher inGroup IIR and Dex(P〈0.05), it is much lower in Dex Group (P 〈0.05) than FR Group. Conclusion Dexmedetomidine canreduce inflammatory reaction by reducing P-STAT3 expression, thus alleviating renal ischemia reperfusion injury andprotecting kidney to some degree.
