基于基因表达谱的肝脏缺血再灌注损伤分子网络构建与关键分子辨识

摘    要:目的基于缺血再灌注小鼠肝脏细胞的基因表达谱构建相关的分子网络,获得高度相关的肝脏缺血再灌注损伤核心网络。方法应用融合表达谱相关性信息的激活子网辨识算法对小鼠肝脏缺血再灌注损伤的基因表达谱进行分析,计算基因表达谱的激活子集,对相关基因子集进行聚类和富集分析。结果与结论肝脏缺血再灌注损伤激活子集分析结果表明,JAK/STAT、MAPK、TLR等信号通路参与该损伤的病理生理过程。利用生物信息学方法对肝脏缺血再灌注损伤表达谱进行数据分析,将为进一步了解肝脏缺血再灌注损伤发病机制提供新的恩路。

Abstract:Objective To construct molecular networks based on ischemia-reperfusion(IR) in gene expression profiles of mouse liver cells, and obtain highly relevant hepatic IR injury in the core network. Methods The gene expression profi ling of mouce liver IR injury was analyzed by integration expression profiling relevant information on activated subset identi fication algorithm. The activation of a subset of the gene expression profiles was calculated. The related gene subset poly classes and enrichment were also explored. Results and Conclusion The results show that JAK/STAT, MAPK and TLR signaling pathways are involved in the pathophysiological process of the injury. Bioinformatics methods provide new ideas about pathogenesis of liver IR injury in mice.
