黄连解毒汤传统饮片汤剂与配方颗粒汤剂中小檗碱的药动学的比较

目的比较黄连解毒汤传统饮片汤剂与配方颗粒汤剂中盐酸小檗碱的药动学行为的一致性。方法采用高效液相色谱法。色谱柱：Diamonsil C18（4.6 mm×250 mm,5μm）;柱温：35℃;流速1 mL.min-1;流动相为甲醇-0.05 mol.L-1KH2PO4-冰醋酸（40∶60∶1,v/v）,检测波长为345 nm。结果灌胃低、中、高剂量组的2种汤剂后,大鼠血浆中小檗碱Cmax分别为：1.952和2.195μg.mL-1,3.188和3.388μg.mL-1,4.10和4.54μg.mL-1;AUC0～∞分别为：10.468和11.841μg.h.mL-1,18.50和20.9μg.h.mL-1,23.16和25.20μg.h.mL-1;tmax均为3.0 h。结论经统计分析,2种汤剂中盐酸小檗碱的Cmax、tmax、AUCo～∞以及药时曲线均无显著差异性,显示小檗碱在2种汤剂中的药动学过程无显著性差异。

Pharmacodynamics of berberine in traditional slice decoction and dispensing granule decoction of Huanglian Jiedu Tang

Objective To compare the pharmacodynamic consistence of berberine between traditional slice decoction and dispensing granule decoction of Huanglian Jiedu Tang.Methods HPLC method was used to detect berberine with Diamonsil C18（4.6 mm×250 mm,5 μm）column.The column temperature was 35 ℃and the flow rate was 1 mL·min-1.The mobile phase consisted of MeOH-0.05 mol·L-1KH2PO4-glacial acetic acid（40∶60∶1）,and the wavelength was 345 nm.Results After administering low,middle,and high dose of the two decoctions,Cmax of berberine was 1.952 and 2.195 μg·mL-1,3.188 and 3.388 μg·mL-1,and 4.10 and 4.54 μg·mL-1 respectively.AUC0～∞ was 10.468 and 11.841 μg·h·mL-1,18.50 and 20.9 μg·h·mL-1,and 23.16 and 25.20 μg·h·mL-1 respectively.and tmax was 3.0 h.Conclusion Cmax,tmax,AUCo～∞ and C-T curves between the two decoctions have no significant difference,indicating that the pharmacodynamics of berberine have no significant difference.
