电针对吗啡戒断大鼠血、脑组织NO/NOS水平的影响

摘    要:目的 :观察电针足三里穴对吗啡戒断大鼠血液 ,脑组织中一氧化氮 (NO)水平和一氧化氮合酶(NOS)活力的影响 ,探讨其对吗啡戒断大鼠调整作用的机理。方法 :给大鼠连续注射递增量吗啡 8d,造成其对吗啡依赖。停药后第 2 d开始给于强度为 2 m A,频率 2~ 1 0 0 Hz混频刺激。NO采用硝酸还原酶的化学比色法测定 ,NOS采用化学比色法测定 ,中枢损毁采用电凝法损毁下丘脑腹内侧核 (ventral medialhypothalamus,VMH)。结果 :模型组大鼠体重与正常组比明显降低 P<0 .0 1 ) ;血清 NO水平升高 (P<0 .0 1 ) ,NOS活力增强 (P<0 .0 5 ) ,脑组织中 NO、NOS水平与正常组比较均无显著差异 ;电针组大鼠体重、血清 NO、NOS含量与正常组相比无显著差异。结论 :电针对吗啡戒断大鼠血清 NO、NOS水平具有良性调整作用 ,损毁下丘脑 VMH后 ,对电针的调整作用有一定影响

Abstract:Objective:To observe effects of electroacupuncture(EA) on nitric oxide(NO) and nitric oxide synthase(NOS) levels in brain and blood of morphine withdrawl rats,and to research the mechanisms of modulation effect in these rats. Method:The Wistar rats were divided into five groups randomly: control group(C), model group(M), model EA group(ME),model center destroy group (MCD) and model center destroy EA group(MCDE). Morphine dependence was induced in Wistar rats by successive injection of morphine for 8 days and EA 2 mA in strength at Zusanli(ST-36) points was given next day after withdrawal of morphine with mix frequencies of 2~100Hz. Result:The weight of model group rats were reduced. The NO and NOS in serum were increased respectively (p<0.01, P<0.05), whereas no significant differences of them in the cerebrum tissue were observed. After EA, there were no significant differences in weight, NO and NOS in serum between normal control group and model group. Conclusion: EA has innocent modulation effect on the contents of NO and NOS in sera in morphine withdrawal rats.
