尾部悬吊对小鼠胸腺细胞凋亡的影响

摘    要:目的:探讨1d和2d尾吊对小鼠胸腺细胞凋亡的影响。方法:采用尾部悬吊小鼠模型模拟失重,碘化丙啶(PI)染色法检测凋亡细胞染色体断裂,Annexin-V分析法检测凋亡的细胞膜的变化。结果:尾吊小鼠未出现明显的染色体断裂,悬吊1d,总凋亡细胞数和早期凋亡细胞数呈加趋势;悬吊2d早期地亡细胞数明显升高,总凋亡细胞数极显著增加,结论:尾吊可能增强的胸腺细胞凋亡。

Abstract:Objective To investigate the effect of simulated weightlessnes s on apoptosis of thymus cells in mice. Method Tail-suspended mice w ere used as an animal model of simulated weightlessness; chromosome breakage wa s measured by PI staining,and changes of cell membrane were determined by Annexi n-V analysis. Result As compared with the control, there was no o bvious chromosome breakage; the total number of apoptotic cells and number of ea rly apoptotic cells increased in 1 d suspended group; the number of early apopto tic cells increased significantly and the total number of apoptotic cells increa sed very significantly in 2d suspended groups. Conclusion These resu lts suggested that simulated weightlessness could promote apoptosis of thymus ce lls.
