贲门癌组织中P53、P16和P21WAF-1蛋白的表达

摘    要:目的:探讨贲门癌组织中肿瘤抑制基因P53、P16和P21^WAF-1蛋白的表达及其临床生物学意义。方法:采用免疫组织化学法检测60例贲门癌组织中P53、P16和P21^WAF-1蛋白的表达。30例正常人贲门粘膜组织作为正常对照。结果:贲门癌组织中P53蛋白的阳性表达率为66.7%,较正常对照组(13.2%)明显增高;贲门癌组织中P16蛋白的阳性表达率为41.6%,较正常对照组(79.2%)降低;贲门癌组织中P21^WAF-1蛋白的阳性表达率为21.6%,较正常对照组(43.2%)降低,差异有统计学意义。贲门癌组织中至少有一项基因表达改变。且多发生2种以上基因表达改变。结论:P53、P16和P21^WAF-1蛋白在贲门癌发生发展中起重要作用。基因改变的累积可能是贲门癌发生的重要分子生物学基础。

Abstract:Aim:To explore the relationship between the expression of P53,P16 and the clinicobiological behavior of human cardiac carcinoma.Methods:The protein expressions of P53,P16, and P21 WAF 1 were detected in specimens from 60 cases of cardiac carcinoma and 30 cases of normal cardiac mucosa tissues with immunohistochemical method.Results:The results showed that the positive rate of P53 protein expression in cardiac carcinoma tissues was significantly higher than that in normal cardiac tissues (66.7% vs 13.2%) while the positive rates of P16 and P21 WAF 1 protein expression were lower than those in normal cardiac tissues (41.6% vs 79.2% for P16 protein and 21.6% vs 43.2% for P21 WAF 1 protein). There was at least one of gene expression alterations in cardiac carcinoma tissues and most of cardiac carcinoma tissues had over two gene expression alterations.Conclusion:P53,P16, and P21 WAF 1 proteins play important roles in the genesis and development of cardiac carcinoma.Accumulation of the gene expression alterations may be the molecular basis for the genesis of cardiac carcinoma.
