反相高效液相色谱法对不同来源西红花药材中西红花苷-1、苷-2的定量分析

 目的：测定西红花药材中西红花苷_-1、苷-2的含量。方法：采用反相高效液相色谱，以Nova-Pak C₁₈(4 μm,150 mm×3.9 mm)柱为色谱柱，甲醇-水（55∶45）为流动相，测定波长为(440±1）nm。结果：西红花苷_-1的平均回收率为99.53%，日内、日间RSD分别为1.0%，1.6%；西红花苷-2的平均回收率为99.43%，日内、日间RSD分别为1.3%，1.8%。进口西红花商品药材中有2种未测出或只有痕量，3种含量较低（西红花苷_-1为80.9～102.0 mg·g_-1生药，西红花苷-2为44.5～62.3 mg·g_-1生药），而国产西红花商品药材中含量最高（西红花苷_-1为176.2 mg·g_-1生药，西红花苷-2为108.5 mg·g_-1生药）。结论：建立的RP-HPLC法测定不同来源西红花药材中西红花苷_-1、苷-2的含量，操作简便，结果准确。

Determination of crocusⅠ and crocusⅡ inCrocus sativus L. from Various sources by RP-HPLC

OBJECTIVE To determine the content of crocusⅠ and crocusⅡ in Crocus sativus L. of domestic and foreign available Crocus sativus L. samples.METHODS CrocusⅠ and crocusⅡ were separated on Nova-Pak C18 column with a mobile phase methanol-water (55∶45)and detection wavelength of 440nm.RESULTS The standard curve of crocusⅠwas linear in the concentration range of 50～300 mg·L_-1 (r=0.9999),the average rate was 99.53%.The intra-day and inter-day RSD were 1.0% and 1.6% respectively. The standard curve of crocus　Ⅱ was linear in the concentration range of 30～300 mg·L_-1(r=0.9999),the average rate was 99.43%. The intra-day and inter-day RSD were 1.3% and 1.8% respectively. The results showed that the imported Crocus sativus L. contained no or a small amount of crocusⅠ and crocusⅡ (crocusⅠ:0～102.0 mg·g_-1; crocusⅡ: 0～62.3 mg·g_-1), but the domestic amounted almost crocusⅠ176.2 mg·g_-1 and crocusⅡ108.5 mg·g_-1.CONCLUSIONS It is a simple and accurate method for the determine of crocusⅠ and crocusⅡ in Crocus sativus L.
