高效液相色谱法测定瑞巴匹特血浆浓度

 目的：建立测定血浆内瑞巴匹特浓度的高效液相色谱法。方法：Kromasil C₁₈色谱柱，甲醇-0.05 mol·L^-1磷酸二氢钾(55∶45，pH3.0)为流动相，并由序贯单纯形法确定此为最佳配比点，流速1.0 ml·min^-1，检测波长230 nm。结果：瑞巴匹特在15～1200 μg·L^-1范围内,血浆药物浓度C与色谱峰高H呈现良好线性相关，血浆中瑞巴匹特平均相对回收率为（97.5±0.5）%(n＝5),平均绝对回收率为（90.3±1.8）%(n＝5)；平均日内和日间RSD分别为4.68%和5.66%(n＝5)；方法对血浆瑞巴匹特最低检出浓度为2 μg·L^-1。结论：本方法适于血浆中瑞巴匹特的测定。

Determination of rebamipide in plasma by high performance liquid chromatography

OBJECTIVE To establish a high performance liquid chromatographic method to determine rebamipide in plasma.METHODS The concentration of the drug was determined within 10 minutes by the method on a column of kromasil C₁₈，using a mobile phase consisted of methanol-potassium di-hydrogen phosphate (55∶45，pH 3.0)and monitored at 230 nm.The rebamipide concentrations in plasma were determined following a single oral dose of 600 mg.RESULTS Good linear relationship between concentration in plasma and peak area in the ranges of 10～1200 μg·L^-1 was obtained．Good precision and reproducibility were found．The relative and absolute average recoveries were (97.5±0.51)% and (90.3±1.78)%(n＝5),respectively．The detection limit was 2 μg·L^-1．CONCLUSIONS The method is suitable for the determination of rebamipide in body fluids．
