摘    要: 目的:建立用高效液相色谱法(HPLC)测定血清中富马酸奎的平浓度的方法。方法:HPLC测定,流动相为甲醇-水-1 mol·L-1醋酸铵-5 mol·L-1氨水(400∶100∶5∶1)的混合液体,pH9.7,流速1 mL·min-1,C18柱,紫外检测波长254 nm,以外标法定量。结果:线性范围10~300 μg·L-1,最低检测限5 μg·L-1,平均提取回收率93.10%,日内与日间精密度平均分别为3.41%和4.16%。结论:用HPLC可以较好的对血清中富马酸奎的平进行定性和定量。

Abstract:OBJECTIVE To establish an HPLC assay method for the determination of quetiapine fumarate level in human serum. METHODS Using a mixture liquid of methanol:water:1 mol·L-1 ammonium acetate:5 mol·L-1 NH4OH(400∶100∶5∶1) as mobile phase, pH9.7, flow rate was 1 mL·min-1, C18 column, the wave length of detection was 254 nm, quetiapine fumarate level was determined by means of external standard method.RESULTS A linearity was obtained from 10~300 μg·L-1 of Quetiapine Fumarate level in serum with a good correlation(r=0.9997). Detection limit was 5 μg·L-1. The average extraction recovery was 93.10%. The cofficients of inter-day and intra-day variations were 3.41% and 4.16% respectively. CONCLUSIONS Using HPLC can be used for the detection of quetiapine fumarate in human serum.
